摘 要:【目的】构建藻株X在不同培养条件下的生长动力学和油脂合成动力学模型,阐释外源添加肌醇提高藻株X油脂产量的生理调节机制。【方法】设置自养培养、异养培养和混养培养3种培养模式,探究其对藻株X生长特性的影响;比较藻株X生长(生物量、葡萄糖浓度和油脂产量)和生理(蛋白质、总糖、丙二醛和超氧阴离子自由基)指标的变化,探究外源添加肌醇对藻株X油脂合成的影响。【结果】构建了不同培养模式的动力学模型(Logistic、Luedeking-Piret以及Luedeking-Piret修正方程),结果表明藻株X的油脂合成是部分生长偶联型。当肌醇添加浓度为2.5 mg/L时其油脂含量达到最大值1.70 g/L,相较于对照组提高了10.22%,外源添加肌醇的处理组与对照组相比丙二醛、超氧阴离子自由基和可溶性蛋白质含量均降低,总糖含量增加。【结论】藻株X的油脂合成是部分生长偶联型且在混养条件下更适合其油脂的积累,肌醇可在一定程度上保护微藻细胞免受外界胁迫因素的毒害作用,促进藻株X的生长及积累油脂。[Objective]The purpose of this study is to construct the growth kinetic model and lipid synthesis kinetic model f microalga Xunder different culture conditions. Meanwhile,the physiological regulation mechanisms under the exogenous addition of inositol conditions improve the lipid production of microalga Xis also elucidated.[Method]Three modes,which were autotrophic culture,heterotrophic culture,and mixotrophic culture,were established to explore their influences on the growth characteristics of microalga X.In order to research the effect of exogenous addition of inositol on the lipid synthesis of the producing algae,the growth(biomass,glucose concentration and lipid production)and physiological(protein,total sugar,malondialdehyde and superoxide anion free radicals)indicators of microalga Xwas also investigated.[Result]Three kinetic models(Logistic,Luedeking Piret and Luedeking-Piret modified equations)were established for different culture conditions,and the results showed that the lipid synthesis of microalga Xwas a partial growth coupling type.In addition,the maximum lipid content of 1.70 g/L were obtained when the addition concentration of inositol was2.5 mg/L,which was 10.22% higher than that of the control group.Compared with the control group,the content of malondialdehyde,superoxide anion free radical,and soluble protein was decreased in the treatment group,and an enhancement of the total sugar content was observed.[Conclusion]The lipid synthesis of microalga Xis a partial growth coupling type.More lipid content would be accumulated under the polyculture conditions.Furthermore,the inositol can protect the microalgae cells when they are under the adverse external stress environment,thus promoting the growth and the lipid production of microalga X.
