摘 要:目的:探讨微小RNA-181a (miR-181a)、TGFβR2在食管癌细胞ECA109中的表达水平及其意义。方法:采用实时荧光定量PCR (RT-qPCR)检测HEEC、ECA109细胞株中miR-181a的表达情况。miR-181a模拟物(miR-181a mimics)、miR-181a抑制物(miR-181a inhibitor)转染食管癌细胞系ECA109。RT-qPCR检测各组miR-181a的表达水平。采用CCK8检测miR-181a对EC109细胞增殖能力的影响。采用RT-qPCR、Western Blot检测TGFβR2mRNA及其蛋白的表达水平。结果:与正常食管上皮细胞株相比,食管癌细胞株ECA109中miR-181a的表达水平明显升高。与阴性对照组相比,在miR-181a mimics组,食管癌细胞ECA109中miR-181a的表达水平明显升高,且细胞增殖能力增强。miR-181a inhibitor组,食管癌细胞ECA109中miR-181a的表达水平明显降低,且细胞增殖能力降低。与阴性对照组相比,miR-181a mimics组,食管癌细胞ECA109中TGFβR2mRNA及其蛋白的表达水平明显降低。miR-181a inhibitor组,食管癌细胞ECA109中TGFβR2mRNA及其蛋白的表达水平明显升高。结论:miR-181a在食管癌细胞ECA109中高表达,通过影响TGFβR2的表达参与食管癌的发生发展。Objective: To investigate the expression levels and significance of microRNA-181a(miR-181a) and TGFβR2 in esophageal cancer cells ECA109. Methods: Real-time quantitative PCR(RT-qPCR) was used to detect the expression of miR-181a in HEEC and ECA109 cell lines. Esophageal cancer cell line ECA109 was transfected with miR-181a mimics and miR-181a inhibitor. RT-q PCR was used to detect the expression levels of miR-181a in each group. The effect of miR-181a on the proliferation ability of EC109 cells was detected by CCK8. The expression levels of TGFβR2 mRNA and protein were detected by RT-qPCR and Western Blot. Results: Compared with normal esophageal epithelial cell lines, the expression level of miR-181a in esophageal cancer cell line ECA109 was significantly increased. Compared with the negative control group, in the miR-181a mimics group, the expression level of miR-181a in esophageal cancer cells ECA109 was significantly increased, and the cell proliferation ability was enhanced. In the miR-181a inhibitor group, the expression level of miR-181a in esophageal cancer cells ECA109 was significantly decreased, and the cell proliferation ability was decreased. Compared with the negative control group, the expression levels of TGFβR2 m RNA and protein in esophageal cancer cells ECA109 in the mi R-181a mimics group were significantly decreased. In the miR-181a inhibitor group, the expression levels of TGFβR2 mRNA and its protein in esophageal cancer cells ECA109 were significantly increased. Conclusion: MiR-181a is highly expressed in esophageal cancer cells ECA109, and participates in the occurrence and development of esophageal cancer by affecting the expression of TGFβR2.
