摘  要:目的:研究骨碎补总黄酮对记忆障碍模型小鼠学习记忆能力及雌激素受体(ER)表达的影响。方法:对亚硝酸钠和东莨菪碱两种记忆障碍模型小鼠给予骨碎补总黄酮进行干预,采用跳台实验观察小鼠的学习及记忆能力;采用HE染色法观察小鼠下丘脑的病理变化,免疫组化法观察小鼠下丘脑中ERα表达。结果:与模型组比较,各给药组小鼠潜伏期显著延长(P<0.01),错误次数显著减少(P<0.01),下丘脑ERα的表达显著增加(P<0.01);HE染色显示,模型组小鼠神经细胞排列得比较紧密,细胞间距变大,细胞核染色较深,核仁较为清晰。结论:骨碎补总黄酮使两种记忆障碍模型小鼠的学习记忆能力得到改善,其作用机制可能通过调节下丘脑区ER表达发挥神经保护作用。Objective:To study the effects of total flavonoids of Drynariae Rhizoma on learning,memory ability and estrogen receptor(ER)expression in mice with memory impairment.Methods:Sodium nitrite and scopolamine were given to mice with two kinds of memory impairment models to intervene with total flavonoids of Drynariae Rhizoma,and the learning and memory abilities of mice were observed by step-down test.HE staining was used to observe the pathological changes in the hypothalamus of mice,and the expression of ERαin the hypothalamus of mice was observed by immunohistochemistry.Results:Compared with the model group,the incubation period of mice in each administration group was significantly prolonged(P<0.01),the number of errors was significantly decreased(P<0.01),and the expression of hypothalamic ERαwas significantly increased(P<0.01).HE staining showed that the nerve cells in the model group were arranged closely,the cell spacing became larger,the nucleus staining was deeper and the nucleolus was clearer.Conclusion:The total flavonoids of Drynariae Rhizoma improved the learning and memory abilities of mice with two kinds of memory impairment models,and its mechanism may play a neuroprotective role by regulating the expression of ER in hypothalamus.
