摘  要:目的探讨Pokemon及其相关基因SF2/ASF、BIN1、eIF4E、p-eIF4E在结肠癌中的表达及其相关性。方法通过手术保存的结肠癌、结肠腺瘤及结肠炎性息肉蜡块切片购自惠州市第三人民医院,采用免疫组化检测Pokemon阳性表达,免疫印迹及PCR检测Pokemon、SF2/ASF、BIN1、eIF4E及p-eIF4E的蛋白及mRNA表达。体外培养结肠癌细胞株HT-29进行Pokemon沉默及Pokemon扩增,观察沉默或增加Pokemon基因对结肠癌HT-29细胞SF2/ASF、BIN1、eIF4E及p-eIF4E水平影响及相关性分析。结果Pokemon在结肠癌中呈现强阳性表达,在结肠腺瘤中呈现阳性表达,在结肠炎性息肉中弱表达;与结肠炎性息肉组织相比,结肠腺瘤组织中Pokemon、SF2/ASF、eIF4E及p-eIF4E表达升高,BIN1降低,组间比较差异有统计学意义(P<0.05);与结肠腺瘤组织相比,结肠癌组织中Pokemon、SF2/ASF、eIF4E及p-eIF4E表达升高,BIN1降低,组间比较差异有统计学意义(P<0.05);Pokemon转染组12 h、24 h及48 h时SF2/ASF及eIF4E/p-eIF4E表达增强,BIN1降低。Pokemon沉默组12 h、24 h及48 h时SF2/ASF及eIF4E/p-eIF4E表达降低,BIN1增加;Pokemon与SF2/ASF、eIF4E呈正相关(R _(SF2/ASF)=0.651,R_(eIF4E)=0.286;P均<0.001),与BIN1呈负相关(R=-0.527,P<0.001)。结论过表达Pokemon通过调控SF2/ASF信号抑制BIN1表达,加快eIF4E磷酸化而促进结肠癌发生发展。Objective To investigate the expression of Pokemon and its related gene SF2/ASF,BIN1,eIF4E and p-eIF4E in colon cancer and its correlation.Methods Surgical preservation of colon cancer,colon adenoma and colonic inflammatory polyps wax slices were purchased from the Third People′s Hospital of Huizhou City,using immunohistochemistry to detect Pokemon positive expression,Western blotting and PCR to detect Pokemon,SF2/ASF,BIN1,eIF4E and p-eIF4E protein and mRNA expressions.Colon cancer cell line HT-29 was cultured in vitro for Pokemon silencing and Pokemon amplification,and silencing or increasing the Pokemon gene was observed on the level of SF2/ASF,BIN1,eIF4E and p-eIF4E of colon cancer HT-29 cells and the correlation analysis.Results Pokemon showed strong positive expression in colon cancer,positive expression in colon adenoma,and weak expression in colonic inflammatory polyps.Compared with colonic inflammatory tissue,the expressions of Pokemon,SF2/ASF,eIF4E and p-eIF4E increased,but BIN1 decreased in colon adenoma tissue,and there was a significant statistical difference(P<0.05).Compared with colon adenoma tissue,the expressions of Pokemon,SF2/ASF,eIF4E and p-eIF4E increased,but BIN1 decreased in colon cancer tissue,and there was a significant statistical difference(P<0.05);the expressions of SF2/ASF and eIF4E/p-eIF4E increased at 12 hours,24 hours and 48 hours in the Pokemon transfection group,and BIN1 decreased,while SF2/ASF and eIF4E/p-eIF4E expressions decreased,and BIN1 increased in the Pokemon silence group;Pokemon was positively correlated with SF2/ASF and eIF4E(R_(SF2/ASF)=0.651,R_(eIF4E)=0.286;all P<0.001),there was a negative correlation with BIN1(R=-0.527,P<0.001).Conclusion Overexpression of Pokemon can inhibit the expression of BIN1 by regulating the SF2/ASF signal,accelerate the phosphorylation of eIF4E,and promote the occurrence and development of colon cancer.
