摘 要:目的探究铁死亡相关lncRNAs在肝细胞癌(hepatocellular carcinoma,HCC)患者中的预后价值及其在肿瘤微环境中的潜在价值。方法从TCGA数据库下载HCC患者相关数据,通过构建铁死亡相关mRNA-lncRNA共表达网络筛选出铁死亡相关lncRNAs。通过单因素Cox回归、LASOO回归和多因素Cox回归构建铁死亡相关lncRNAs预后模型和风险评分体系。结果获得由5个铁死亡相关lncRNAs组成的预后模型,包括:AL049840.4、ZFPM2-AS1、AC074117.1、AC099850.3和AL031985.3。风险评分和TNM分期的列线图显示出该模型具有较好的预测能力。KEGG通路结果显示高风险组富集于经典的肿瘤相关通路。该预后模型与肿瘤免疫细胞浸润、免疫功能及免疫检查点基因的表达有关。结论本研究所构建的铁死亡相关lncRNAs预后模型对预测HCC患者生存预后具有一定的准确性和可靠性,为铁死亡和HCC免疫之间的相互作用提供了线索。靶向铁死亡联合免疫检查点抑制剂有望为HCC治疗提供新的视角。Objective To explore the prognostic value of ferroptosis-related lncRNAs in hepatocellular carcinoma(HCC)patients and their potential value in tumor microenvironment.Methods The data of HCC patients were downloaded from TCGA database,and ferroptosis-related lncRNAs were screened out by constructing ferroptosis-related mRNA-lncRNA co-expression network.The prognosis model and risk scoring system of lncRNAs related to ferroptosis were constructed by univariate Cox regression,LASOO regression and multivariate Cox regression.Results A prognostic model consisting of 5 ferroptosis-associated lncRNAs was established,including AL049840.4,ZFPM2-AS1,AC074117.1,AC099850.3 and AL031985.3.The risk score and TNM staging diagram showed that the model had good predictive ability.KEGG pathway results showed that the high-risk group was enriched in the classical tumor-related pathways.The prognostic model was related to tumor immune cell infiltration,immune function and immune checkpoint gene expression.Conclusion The ferroptosis-related lncRNAs prognostic model constructed in this study has certain accuracy and reliability in predicting the survival prognosis of HCC patients,providing clues for the interaction between ferroptosis and HCC immunity.Targeting ferroptosis combined with immune checkpoint inhibitors is expected to provide a new perspective for the treatment of HCC.
