摘 要:目的探讨核因子E2相关因子2(nuclear factor-erythroid-2-related factor 2,Nrf2)和血红素加氧酶-1(heme oxygenase-1,HO-1)在蓝光损伤大鼠视网膜组织的表达情况及机制方法将40只健康清洁级SD大鼠随机分为对照组与实验A组、B组、C组(n=10),对照组正常饲养,不予以蓝光照射;实验组每天接受2500 Lux的蓝光(峰值450 nm)照射12 h,A组、B组、C组分别照射10 d、20 d、30 d。结束后取大鼠眼球进行HE染色;TUNEL法检测视网膜组织细胞凋亡指数,免疫荧光染色法检测Nrf2、HO-1蛋白表达。结果与对照组相比,随着蓝光暴露时间的增加,实验组视网膜厚度显著下降,尤其是外核层,且光感受器细胞凋亡指数显著增加,视网膜组织内Nrf2显著增加,HO-1表达增加。结论过度暴露于短波蓝光导致视网膜组织厚度下降,主要损伤视锥视杆细胞,激活视网膜组织氧化应激反应,使Nrf2、HO-1表达增加。Objective To investigate the expression and mechanism of nuclear factor-erythroid-2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)in retinal tissue of rats injured by blue light.Methods 40 healthy clean SD rats were randomly divided into control group and experimental group A,B and C(n=10 each).The control group was treated without blue light,and the experimental group was exposed to blue light(peak 450nm)of 2500 Lux for 12 hours every day for 10 days,20 days and 30 days respectively.Afterwards,the eyeballs of rats isolated and stained with HE,the apoptosis index of retinal tissue was detected by TUNEL method,and the expression of Nrf2 and HO-1 protein was detected by immunofluorescence staining.Results Compared with the control group,with the increase of blue light exposure time,the retinal thickness of the experimental group decreased significantly,especially in the outer nuclear layer,and the apoptosis index of photoreceptor cells increased significantly,Nrf2 and HO-1 expression increased significantly.Conclusion Excessive exposure to shortwave blue light can decrease the thickness of retinal tissue,mainly damage the cone rod cells,activate the oxidative stress response of retinal tissue,and increase the expression of Nrf2 and HO-1.
