摘  要:目的探讨肌电相同的喉内肌与喉外肌的乙酰胆碱(Ach)、乙酰胆碱受体(nAChR)、Na^(+)-K^(+)-ATP酶的浓度,运动终板(MEP)分布,以及肌纤维类型的相似性,为精准选择相应喉外肌支配的神经进行神经或神经肌蒂移植治疗声带麻痹提供参考。方法10只健康成年犬,取环杓侧肌及与其肌电相同的下颌舌骨肌、甲状舌骨肌,环杓后肌及与其肌电相同的颏舌骨肌、胸骨甲状肌;应用ELISA法测量各肌肉中Ach、AChR、Na^(+)-K^(+)-ATP酶浓度;用乙酰胆碱酯酶染色观察各肌肉中运动终板的数量;用ATP酶染色比较各肌纤维中慢肌纤维(MHC-Ⅰ)和快肌纤维(MHC-Ⅱ)个数及横截面积比,用荧光定量PCR检测nAChR、MHC-Ⅰ、MHC-ⅡmRNA的表达。结果①下颌舌骨肌Ach浓度低于环杓侧肌(P<0.05),甲状舌骨肌与环杓侧肌差异无统计意义(P>0.05);下颌舌骨肌Na^(+)-K^(+)-ATP酶浓度高于环杓侧肌(P<0.05),甲状舌骨肌与环杓侧肌差异无统计学意义(P>0.05);胸骨甲状肌Ach浓度低于环杓后肌(P<0.05),颏舌骨肌与环杓后肌比较差异无统计学意义(P>0.05)。胸骨甲状肌Na^(+)-K^(+)-ATP酶浓度高于环杓后肌(P<0.05),颏舌骨肌与环杓后肌差异无统计学意义(P>0.05)。下颌舌骨肌、甲状舌骨肌与环杓侧肌之间及胸骨甲状肌、颏舌骨肌与环杓后肌nAChR浓度比较差异均无统计学意义(P>0.05)。②下颌舌骨肌和甲状舌骨肌的运动终板数量与环杓侧肌比较差异均无统计学意义(P>0.05),胸骨甲状肌和颏舌骨肌与环杓后肌比较差异亦无统计学意义(P>0.05)。③下颌舌骨肌和甲状舌骨肌的MHC-Ⅱ/MHC-Ⅰ个数比与环杓侧肌比较均无显著差异(均为P>0.05),下颌舌骨肌和甲状舌骨肌的MHC-Ⅱ/MHC-Ⅰ横截面积比与环杓侧肌比较差异均有统计学意义(均为P<0.05)。颏舌骨肌的MHC-Ⅱ/MHC-Ⅰ个数比和横截面积比与环杓后肌比较差异均有统计学意义(均为P<0.05);胸骨甲状肌的MHC-Ⅱ/MHC-Ⅰ个数比与环�Objective To study the similarity of the acetylcholine(Ach),acetylcholine receptor(nAChR),Na^(+)-K^(+)-ATP enzyme concentration,the distribution of the motor end-plate(MEP)and the types of muscle fibers between the internal and external laryngeal muscles with the same EMG,and to provides a reference for accurately selecting the nerves innervated by the corresponding external laryngeal muscles and for the treatment of vocal cord paralysis with nerve or neuromuscular pedicle transplantation.Methods The mylohyoid muscles(MM),geniohyoid muscles(GM)、sternothyroid muscles(SM)、thyrohyoid muscles(TM)、lateral cricoaryteoid muscle(LCA)and posterior cricoaryteoid muscle(PCA)were obtained from ten healthy adult dogs.ELISA was used to measure the concentration of Ach、nAChR、Na^(+)-K^(+)-ATPase;acetylcholinesterase staining was used to observe the number and area of motor end-plates,and ATPase staining was used to compare the types of muscle fibers in each group;fluorescence quantitative PCR was used to detect the expression of nAChR、MHC-Ⅰ、MHC-ⅡmRNA.Results①Ach concentration:MH was lower than LCA(P<0.05),but there was no significant difference between TH and LCA(P>0.05);Na^(+)-K^(+)-ATPase concentration:MH was higher than LCA(P<0.05),and there was no significant difference between TH and LCA(P>0.05).ST’Ach concentration was lower than PCA,there was no significant difference between GH and PCA(P>0.05);the Na^(+)-K^(+)-ATPase concentration of ST was higher than PCA(P<0.05),there was no significant difference between GH and PCA(P>0.05).There was no significant difference between MH、TH and LCA in nACh R concentration,so were ST,GH and PCA.②There was no significant difference between MH、TH and LCA in MEP number,so were ST,GH and PCA(P>0.05).③There was no significant difference between MH、TH and LCA in the number ratio of MHC-Ⅱ/MHC-Ⅰ(P>0.05),while there was significant difference in the cross-sectional area ratio of MHC-Ⅱ/MHC-Ⅰ(P<0.05).There was significant difference between GH and PCA in the
