摘　　要：目的:探讨YAP1蛋白对人胚绒毛癌细胞(JAR)增殖、迁移与侵袭能力的调控作用及其与稽留流产发病的可能机制.方法:收集2018年7月在绍兴市妇幼保健院确诊为稽留流产的患者4例为实验组,选取同期自愿要求行人工流产的正常早孕妇女4例为对照组,通过RT-qPCR和Western Blot验证稽留流产绒毛组织中YAP1的表达水平.用siRNA干扰下调JAR细胞中YAP1的表达(si-YAP1),采用RTCA监测其增殖、迁移和侵袭能力的变化.Western Blot法分析YAP1蛋白表达下调后的细胞中凋亡相关蛋白、胚胎植入相关蛋白表达.结果:与对照组比较,YAP1在稽留流产患者绒毛组织中的表达上调;YAP1表达降低显著抑制了JAR细胞增殖、迁移和侵袭能力,凋亡相关蛋白BAX表达上调,胚胎植入相关蛋白PI3K和Akt表达下调,IRS1表达上调.结论:YAP1的异常表达很可能通过调控凋亡发生和影响胚胎植入相关信号通路来参与稽留流产的发病过程.Objective:To investigate the role of Yes-associated protein 1(YAP1)in regulating proliferation,migration,and invasion of human choriocarcinoma cell line(JAR)and its possible role in missed abortions;Methods:In January 2018,four patients with missed abortion who were diagnosed at Shaoxing Maternal and Child Health Hospital were enrolled in the experimental group,and four healthy early-pregnant women willingly desired abortions during the same time.The expression of YAP1 in the chorionic villus tissue of missed abortion was assessed using RT-qPCR and Western blot.The expression of YAP1 in transfected cells was inhibited using siRNA interference.Transfected cells were examined for YAP1 expression using RT-qPCR and Western blotting analyses.The impact of YAP1 knockdown on cell proliferation,migration,and invasion was examined using RTCA.The protein expression levels of apoptosis-related proteins and signaling pathways connected to proteins related to embryo implantation were examined using Western blot.Results:In the chorionic villus tissues of the missed abortion group,the YAP1 expression is up-regulated in comparison to the control group;the decreased YAP1 expression(si-YAP1)dramatically hinders JAR cells’ability to proliferate,migrate,and invade.In the si-YAP1 group,the protein expression levels of the embryo implantation-related protein IRS1 are up-regulated,the protein expression levels of PI3K and Akt are down-regulated,and the protein expression levels of the apoptosis-related protein BAX are elevated.Conclusions:Abnormal expression of YAP1 may be connected to the etiology of missed abortion by regulating apoptosis and affecting the signaling pathways related to embryo implantation.
