二氢杨梅素对小鼠淋巴瘤L5178Y细胞TK基因的致突变作用

摘  要:用小鼠淋巴瘤L5178Y细胞TK基因的致突变实验来评价二氢杨梅素的致突变性。在代谢活化和非代谢活化条件下,8种不同质量浓度的二氢杨梅素,作用在一定数量的含有小鼠淋巴瘤L5178Y细胞中,作用3 h后,离心将上清液弃去加入完全DMEM培养液,连续培养48 h后,分别铺PE_(2)平板效率和MF突变频率的平板于96孔板中,显微镜下观察MF中的突变细胞集落,计数各剂量组突变细胞的集落数,比较大小集落数与阴性对照组,进行数据统计分析。在代谢活化和非代谢活化条件下的二氢杨梅素均未引起该细胞突变率的改变。因此,当二氢杨梅素的终质量浓度为39.06~5000μg/mL时,未能引起小鼠淋巴瘤细胞TK基因位点的致突变性作用。To evaluate the L5178Y TK gene mutation in mouse lymphoma cells induced by dihydromyricetin,L5178Y cells were treated for 3 h with eight different concentrations of dihydromyricetin in the absence and presence of metabolic activation system.The supernatant was discarded,and the DMEM media was added to culture for 48 h.The PE_(2) plate efficiency and MF mutaion freequency were be pared in plate respectively.The mutant cell colonies in the MF were observed under a microscope;the number of mutant cell colonies in each dose group was counted;and the number of large and small colonies was compared with the negative control group for statistical analysis.The mutation rate of this cell wasn t changed by dihydromyricetin under metabolic.Therefore,the final concentration of dihydromyricetin from 39.06μg/mL to 5000μg/mL failed to cause the mutagenicity of TK gene locus in mouse lymphoma cells.
