野生彩茶“蓝宝石”抗褐化及丛生芽诱导研究

摘　　要：[目的]建立高效的不定芽离体再生体系是野生彩茶商业化快速生产的关键技术。[方法]以野生彩茶“蓝宝石”叶片为外植体试材,探究不同激素、浓度及处理对其外植体灭菌、抗褐化、不定芽分化、丛生芽增殖的影响。[结果]野生彩茶“蓝宝石”腋芽诱导不定芽最佳培养基为MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+IBA 1.0 mg/L,丛生芽继代增殖最佳培养基为MS+6-BA 2.0 mg/L+NAA 1.0 mg/L+IBA 2.0 mg/L。不定芽增殖倍数达到7倍,增殖苗形态健壮,生长快,35 d可继代1次。[结论]该研究可为珍贵彩茶树快速繁殖打下基础。[Objective]To establish an efficient in vitro regeneration system of wild color tea Camellia sinensis cv.“lanbaoshi”.[Method]The leaves of color tea tree lanbaoshi were used as explants to explore the effects of different stimulants,concentrations and treatments on explants sterilization,browning resistance,adventitious bud differentiation and cluster bud proliferation.[Result]The results showed that the best medium for inducing adventitious buds from axillary buds of wild colorful tea lanbaoshi was MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+IBA 1.0 mg/L,the best medium for subculture and proliferation of cluster buds was MS+6-BA 2.0 mg/L+NAA 1.0 mg/L+IBA 2.0 mg/L.The multiplication multiple of adventitious buds reached 7 times.The proliferating seedlings were robust and grew fast,they could be subcultured once in 35 days.[Conclusion]This study can lay a foundation for the rapid propagation of precious colorful tea.
