摘 要:目的 检测B慢性淋巴生物细胞白血病/淋巴瘤11A遗传物质基因(B-cell chronic lymphocytic leukemia/immunocytoma,BCL11A)和乳腺癌表达相互之间已有的关联性,并对其表达与乳腺癌发生和进展之间的内在关系进行探究,总结病理意义。方法 (1)研究对象是福建省医科大学附属协和医院在2013年1月—2020年12月,收治的乳腺癌患者122例,所有患者均进行了手术治疗,所有患者都是女性,年龄29~83岁,中位年龄48岁,收集患者的病理学相关资料。根据其ER、PR、Her-2生物表达的不同展开分子分型。采用实时荧光定量聚合酶链反应技术检测44例乳腺癌生物组织,与16例癌旁生物组织中BCL 11A mRNA的基因表达实际状况。(2)应用免疫组织化学染色法检测在RT-qPCR实验中所运用的44例乳腺癌生物组织,与16例癌旁生物组织中BCL 11A蛋白的基因表达实际状况,分析BCL 11A蛋白和BCL11A mRNA遗传表达的关联性。与此同时,除此之外检测乳腺癌病例生物个体组织78例,在这其中管腔A型6例、管腔B型7例、三阴性型56例和Her-2过表达型9例,探究分析BCL 11A蛋白和乳腺癌病理分子分种类型。结果 (1)在乳腺癌及癌旁组织者中,三阴性乳腺癌的BCL 11A mRNA生物表达水平明显超过其他分型,且显著高于癌旁组织,经对照发现其中差异有统计学意义(P<0.05)。(2)BCL 11A蛋白的基因表达和mRNA的基因表达相关,BCL 11A mRNA表达高者,其BCL 11A蛋白表达也高(P <0.05)。(3)免疫组化表明,BCL 11A蛋白在乳腺癌病例中表现为核和浆的阳性表达,同时在三阴性乳腺癌(41/67)里,其生物特征遗传表达,显著性超过了管腔A型(3/17)、管腔B型(3/18)和Her-2过表达型(4/20),其差异有统计学意义(P <0.05)。癌旁组织仅表现为细胞膜的表达。(4)BCL 11A分子的基因表达水平和多重影响因素相关,包含病理组织学分级、是不是产生淋巴结转移、分子分型、ER表达情况、PR表达情况和HER-2表达�Objective To detect the existing correlation between B-cell chronic lymphocytic leukemia/immunocytoma(BCL 11A) and the expression of breast cancer, and to detect the relationship between its expression and the occurrence and progression of breast cancer, and the pathological significance was summarized. Methods(1) The research subjects were 122 breast cancer patients admitted to Fujian Medical University Union Hospital from January 2013 to December 2020. All patients underwent surgical treatment. Real-time fluorescence quantitative polymerase chain reaction technology was used to detect the actual state of BCL 11A mRNA gene expression in 44 cases of breast cancer biological tissues and 16 cases of adjacent biological tissues.(2) Immunohistochemical staining was used to detect the actual gene expression of BCL 11A protein in 44 cases of breast cancer biological tissues and 16 cases of adjacent biological tissues used in RT-qPCR experiments, and the genetic expression of BCL 11A protein and BCL 11A mRNA was analyzed correlation. At the same time, in addition to this, 78 biological individual tissues of breast cancer cases were detected, including 6 cases of luminal A type,7 cases of luminal B type, 56 cases of triple negative type and 9 cases of Her-2 overexpression type.
