摘  要:目的 研究X射线交错互补修复基因1(XRCC1)多态性及EB病毒(EBV)感染与鼻咽癌(NPC)易感性的关系,为临床筛查、预防和诊治提供参考依据。方法 选取2018年4月-2021年4月济南市人民医院97例初治NPC患者和92名健康体检人群分别为NPC组和对照组,比较两组EBV-DNA水平,分析XRCC1基因Arg194 Arg/Trp、Arg280 Arg/His和Arg399 Arg/Gln多态性。结果 NPC组吸烟史、家族史和EBV-DNA阳性占比均高于对照组(P<0.05),NPC组和对照组XRCC1基因Arg194 Arg/Trp、Arg280 Arg/His和Arg399 Arg/Gln位点基因型分布均满足Hardy-Weinber平衡定律(P>0.05),且NPC组Arg194 Trp/Trp基因型和Trp等位基因分布频率分别为(6.19%和31.44%)低于对照组,Arg399 Gln/Gln基因型和Gln等位基因分布频率分别为(28.87%和52.58%)高于对照组,比较差异均有统计学意义(P<0.05)。结论 XRCC1基因多态性和EBV感染可能导致NPC发病风险升高。OBJECTIVE To explore the correlation between X-ray crosscomplementing repair gene 1(XRCC1) polymorphism and Epstein-Barr virus(EBV) infection and susceptibility to nasopharyngeal carcinoma(NPC) so as to provide guidance for clinical screening, prevention, diagnosis and treatment. METHODS From Apr 2018 to Apr 2021, totally 97 patients with NPC who were initially treated in Jinan City People′s Hospital were assigned as the NPC group, and 92 healthy people who received physical examination were assigned as the control group. The EBV-DNA level was compared between the two groups, and the polymorphisms of XRCC1 gene Arg194 Arg/Trp, Arg280 Arg/His and Arg399 Arg/Gln were observed. RESULTS The proportions of the patients with smoking history, family history and positive EBV-DNA of the NPC group were significantly higher than those of the people in the control group(P<0.05). The distribution of the genotypes at XRCC1 gene Arg194 Arg/Trp, Arg280 Arg/His and Arg399 Arg/Gln loci of both the NPC group and the control group met the Hardy-Weinber balance law(P>0.05), and the frequencies of Arg194 Trp/Trp genotype and allele Trp of the NPC group were respectively 6.19% and 31.44%, significantly lower thanthose of the control group, the frequencies of Arg399 Gln/Gln genotype and allele Gln of the NPC group were respectively 28.87% and 52.58%, significantly higher than those of the control group(P<0.05). CONCLUSION The XRCC1 gene polymorphism and EBV infection may result in the increase of risk of NPC.
