摘  要:目的探讨EB病毒感染对甲状腺乳头状癌(PTC)患者三叶因子3(TFF3)和细胞外调节蛋白激酶(ERK)信号通路的影响。方法选取2019年4月-2021年4月于海口市中医医院行甲状腺癌根治术的118例PTC患者为研究组,另选取同期行甲状腺全切/次切手术的甲状腺腺瘤患者42例为对照组,采集两组甲状腺肿瘤标本,应用原位免疫杂交法检测EB基因编码的核糖核酸(EBER-1)表达,免疫组化检测PTC组织和腺瘤组织中TFF3和ERK信号通路相关蛋白表达,并利用图像分析软件测定平均光密度(AOD)。结果研究组患者EBER-1、TFF3和ERK的阳性率分别为75.42%(89/118)、83.90%(99/118)、69.49%(82/118),高于对照组,差异有统计学意义(P<0.05)。研究组患者甲状腺组织TFF3和ERK蛋白AOD值分别为(0.50±0.02)和(0.41±0.01)均高于对照组,差异有统计学意义(P<0.05)。结论EB病毒感染激活TFF3和ERK信号通路,可能参与PTC的发生与发展,TFF3和ERK蛋白可能成为判断PTC病情进展的指标。OBJECTIVE To investigate the effects of EB virus infection on the signaling pathways of trefoil factor 3(TFF3)and extracellular regulatory protein kinase(ERK)in patients with papillary thyroid carcinoma(PTC).METHODS A total of 118 patients with PTC undergoing radical thyroid cancer surgery in the Haikou Traditional Chinese Medicine Hospital from Apr.2019 to Apr.2021 were selected as the study group,and 42 patients with thyroid adenoma undergoing total/sub-total thyroidectomy during the same period were selected as the control group.The specimens of thyroid tumors in both groups were collected to detect the expression of EB gene-encoded ribonucleic acid(EBER-1)by in situ immunohybridization.The expressions of TFF3 and ERK signaling pathway related proteins in PTC tissues and adenoma tissues were detected by immunohistochemistry,and average optical density(AOD)was determined using image analysis software.RESULTS The positive rate of EBER-1,TFF3 and ERK in study group were 75.42%(89/118),83.90%(99/118),and 69.49%(82/118),respectively,which were significantly higher than those in the control group(P<0.05).The AOD values of TFF3 and ERK proteins in thyroid tissues of the study group were(0.50±0.02)and(0.13±0.05),respectively,significantly higher than those in the control group(P<0.05).CONCLUSION EB virus infection may be involved in the occurrence and development of PTC by activating TFF3 and ERK signaling pathways.TFF3 and ERK proteins may be the potential indicators for determining the progression of PTC.
