食管癌组织中miR-34a与survivin的表达及与病理特征的相关性

摘　　要：目的 分析食管癌组织中微小RNA(miR)-34a与生存素(survivin)的表达及与病理特征的相关性。方法回顾性选取2020年10月至2021年10月在新疆医科大学附属肿瘤医院保存的140份手术切除的食管癌组织及癌旁组织标本作为研究资料,对组织标本中miR-34a、survivin表达情况及患者的临床病理特征进行分析。结果 食管癌组织miR-34a相对表达量低于癌旁组织,survivin表达阳性率及表达强度分布均高于癌旁组织,差异均有统计学意义(P<0.05)。Spearman等级相关分析结果显示,食管癌组织miR-34a与survivin表达强度呈负相关(P<0.05)。肿瘤直径≥4 cm、低分化或未分化、临床分期Ⅲ~Ⅳ期、出现淋巴结转移患者癌组织miR-34a相对表达量均低于肿瘤直径<4 cm、高分化或中分化、临床分期Ⅰ~Ⅱ期、未出现淋巴结转移患者,survivin表达阳性率均高于肿瘤直径<4 cm、高分化或中分化、临床分期Ⅰ~Ⅱ期、未出现淋巴结转移患者,差异均有统计学意义(P<0.05)。结论 食管癌组织中存在着miR-34a相对表达量降低和survivin表达上调,两者呈负相关,且均与肿瘤的病理特征具有相关性,可能是促进食管癌发生和进展的机制之一,有望作为临床治疗靶点。Objective To analyze the expression of microRNA(miR)-34a and survivin in esophageal carcinoma and their correlation with pathological characteristics.Methods A total of 140 specimens of surgically resected esophageal carcinoma tissues and adjacent tissues stored in the Affiliated Cancer Hospital of Xinjiang Medical University from October 2020 to October 2021 were selected as the research data.The expression of miR-34a and survivin in tissue samples and the clinicopathological characteristics of patients were analyzed.Results The relative expression of miR-34a in esophageal carcinoma tissues was lower than that in adjacent tissues,and the positive rate and distribution of survivin expression intensity were higher than those in adjacent tissues,the differences were statistically significant(P<0.05).Spearman rank correlation analysis showed that there was a negative correlation between miR-34a and survivin expression intensity in esophageal carcinoma(P<0.05).The relative expressions of miR-34a in tumor tissues of patients with tumor diameter≥4 cm,poorly differentiated or undifferentiated,clinical stageⅢ~Ⅳand lymph node metastasis were lower than those of patients with tumor diameter<4 cm,highly differentiated or moderately differentiated,clinical stageⅠ~Ⅱand no lymph node metastasis.The positive rates of survivin expression were higher than those of patients with tumor diameter<4 cm,highly differentiated or moderately differentiated,clinical stageⅠ~Ⅱand no lymph node metastasis.The differences were statistically significant(P<0.05).Conclusion There is a decrease in the relative expression of miR-34a and an increase in the expression of survivin in esophageal carcinoma,and there is a negatively correlation with each other,which are correlated with the pathological characteristics of the tumor.They may be one of the mechanisms to promote the occurrence and progression of esophageal carcinoma and are expected to be used as a target of clinical treatment.
