摘 要:运用平板涂布法和基因测序结合对舟山地区部分河道的菌株进行分离提纯并进行基因测序,以研究各河道菌株主要分布及组成。调查发现:城关河、城西河、洋岙河、庆丰河、蓬莱河、环南河、解放河、盛家塘等8处河道中各分离出10、7、7、3、5、6、5、4种菌株,合计共47种菌株,其中共有6种特征菌株,其基因测序结果分别为金黄杆菌(Chryseobacterium cucumeris sp. strain)(1-8)、三价砷氧化细菌(Acidovorax sp. strain)(2-1)、寡养单胞菌(Stenotrophomonas sp. strain)(3-3)、假单胞菌(Pseudomonas sp. strain) P41(3-4)、苍白杆菌(Ochrobactrum sp. strain) JZ28(5-5)、根瘤菌(Rhizobium sp. strain) SCAU410(8-3)。其中各典型菌株的菌落特征、细菌形貌及革兰氏染色结果与各细菌特征一致,表明检测结果可信度较高,可作为舟山市周遭河道治理的实验依据。The strains from different rivers in Zhoushan area were isolated,purified and sequenced by plate coating method and gene sequencing,so as to study the main distribution and composition of strains in each river. The investigation found that 10,7,7,3,5,6,5 and 4 strains were isolated from 8 river courses,including Chengguan River,Chengxi River,Yang’ao River,Qingfeng River,Penglai River,Huannan River,Jiefang River and Shengjiatang. A total of 47 strains were isolated,including six characteristic strains. The gene sequencing results were Chrysobacterium sp. strain(1-8),Acidovorax sp. strain(2-1),Stenotrophomonas sp. strain(3-3),Pseudomonas sp. strain P41(3-4),Ochrobactrum sp. strain JZ28(5-6),Rhizobium sp. strain SCAU410(8-3). Among them,the colony characteristics,bacterial morphology and Gram staining results of each typical strain are consistent with those of each bacteria,indicating that the detection results are highly reliable and can be used as experimental basis for river regulation around Zhoushan City.
