摘 要:目的:探究高危型人乳头瘤病毒(HPV)感染及SLC5A8基因、RASSF2A基因启动子甲基化在宫颈病变中的变化。方法:收集本院2018年5月-2021年5月收治的186例宫颈病变患者临床资料,根据阴道镜宫颈活检病理诊断结果,分为宫颈上皮内瘤变I级组(n=73)、宫颈上皮内瘤变II级组(n=57)和III级组(n=56),同时收集正常细胞43份作为对照组(n=43)。检测4组标本HPV DNA、SLC5A8以及RASSF2A基因启动子甲基化阳性率。结果:对照组、宫颈上皮内瘤变I级、II级、III级组标本HPV DNA(0、28.8%、63.2%、92.9%)、SLC5A8(0、30.1%、66.7%、89.3%)以及RASSF2A(0、13.7%、49.1%、82.1%)基因启动子甲基化阳性率依次升高(均P<0.05)。结论:高危型HPV感染及SLC5A8、RASSF2A基因启动子甲基化在宫颈病变组织中异常升高,并随病情发展而增加。Objective: To investigate the situation of high-risk human papilloma virus(HPV) infection, and to study positive rate of methylation of SLC5 A8 and RASSF2 A promoter in the cells of cervical lesions. Methods: The clinical data of 186 patients with cervical diseases from May 2018 to May 2021 were collected. These patients were divided 73 cases with cervical intraepithelial neoplasia grade I in group A, 57 cases with cervical intraepithelial neoplasia grade II in group B, and 56 cases with cervical intraepithelial neoplasia grade III in group C according to their pathological diagnosis results of cervical biopsy under colposcopy. Meantime, another 43 women with normal cervical cells were included in control group. The positive rates of HPV DNA, SLC5 A8, and RASSF2 A gene promoter methylation in the cervical specimens of the patients in the four groups were detected. Results: The positive rates of DNA in the cervical specimens of the patients in the control group, group A, group B, and group C were 0, 28.8%, 63.2%, and 92.9%, respectively, the positive rates of SLC5 A8 in the cervical specimens of the patients in the control group, group A, group B, and group C were 0, 30.1%, 66.7%, and 89.3%, respectively, and the positive rates of RASSF2 A gene promoter methylation in the cervical specimens of the patients in the control group, group A, group B, and group C were 0, 13.7%, 49.1%, and 82.1%, respectively, which all had increased gradually(all P<0.05). Conclusion: The rates of high-risk HPV infection and SLC5 A8, and RASSF2 A gene promoter methylation in the cervical specimens of the patients with cervical lesions are increase abnormally, and which will increase with the progression of the cervical lesions.
