摘 要:尽管啮齿动物等腹膜纤维化模型为腹膜纤维化的发生、发展机制提供了部分信息。遗憾的是,目前尚无与人类具有高度生理学、解剖学相似性的大型动物模型,限制了动物模型在临床治疗效果评估中的应用。合适的腹膜纤维化动物模型,对于纤维化的研究和抗纤维化新药研发等具有重要意义,而实验动物、造模方法、评价指标的合理选择与匹配,是评价其科学性和有效性的关键。通过系统查阅PubMed、中国知网(CNKI)等数据库中的腹膜纤维化研究文献,从实验动物选择、造模原理和方法、特点、评价指标及其在中医药中的应用等方面,总结了近年来常用的腹膜纤维化动物模型的研究进展。常用的腹膜纤维化动物模型造模方法,是在尿毒症模型和非尿毒症模型的基础上进行的,包括生物诱导模型、化学诱导模型和联合模型等。与生物诱导模型、联合模型相比较,化学诱导模型利用生物不相容性PDF、利用药物如CG、MGO、LPS等形成对腹膜持续的刺激,腹膜形态、功能改变显著,且无需手术操作简单,造模时间较短,在国内外的应用更为广泛。可为腹膜纤维化的发生机制、抗纤维化药物的筛选等研究中的动物模型选择提供参考。Although rodents and other models of peritoneal fibrosis provide some information for the occurrence and development of peritoneal fibrosis.Unfortunately,there is no large animal model with high physiological and anatomical similarity with human,which limits the application of animal model in the evaluation of clinical treatment effect.A suitable animal model of peritoneal fibrosis is of great significance for the study of fibrosis and the research and development of new anti-fibrosis drugs,and the reasonable selection and matching of experimental animals,modeling methods and evaluation indicators are the key to evaluate its science and effectiveness.By systematically consulting the research literature of peritoneal fibrosis in PubMed,CNKI and other databases,this paper summarized the research progress of commonly-used animal models of peritoneal fibrosis in recent years from the aspects of selection of experimental animals,principles and methods of modeling,characteristics,evaluation indexes and their application in traditional Chinese medicine.The commonly-used animal models of peritoneal fibrosis are based on uremic model and non-uremic model,including biological induction model,chemical induction model and combined model and so on.Compared with the biological induction model and the combined model,the chemical induction model uses bioincompatible peritoneal dialysis fluid(PDF)and drugs such as chlorhexidine gluconate(CG),methyl glyoxal(MGO)and lipopolysaccharide(LPS)to stimulate the peritoneum continuously,and the morphology and function of the peritoneum change significantly.The operation is simple,and the modeling time is shorter,and so it is more widely used at home and abroad.This paper can provide a reference for the selection of animal models in the study of the mechanism of peritoneal fibrosis and the screening of antifibrotic drugs.
