摘  要:目的:研究豪猪刺总氨基酸的最佳醇提工艺。方法:采用HPLC-柱前衍生化法测定豪猪刺中氨基酸类12个化学成分,并以总氨基酸量为评价指标,乙醇体积分数、液料比、提取时间和样品粒度为影响因素,通过单因素及Box-Behnken试验优化豪猪刺醇提工艺。结果:优化得到的豪猪刺的最佳醇提工艺为:样品过4号筛,加液料比17∶1(mL/g)、浓度为76%的乙醇提取2 h,在此条件下总氨基酸提取率为3.47%。结论:本该实验优选的醇提工艺稳定、合理可行,可为豪猪刺有效部位研究提供参考。Objective:To optimize the extraction process parameters of total amino acids from porcupine spines.Methods:The ethanol extraction process of porcupine spines was optimized by single factor and Box-Behnken test with total amino acid content as evaluation index and ethanol concentration,liquid-solid ratio,extraction time and sample size as influence factors.Results:The optimal extraction process was as follows:the sample passed through sieve NO.4,the ratio of liquid to solid was 17∶1(mL/g),and the concentration of 76%ethanol was extracted for 2 h.Under these conditions,the total extraction rate of amino acids was 3.47%.Conclusion:The optimized ethanol extraction process is stable,reasonable and feasible.The method is simple and reliable,and can provide a basis for the subsequent study of effective parts of porcupine spines.
