响应面法优化天名精总黄酮提取工艺研究

摘  要:本实验以天名精为原料,通过响应面法优化天名精总黄酮进行提取工艺研究。选择酶量、液料比、超声时间、酶解时间和酶解温度等因素分别考察其对天名精总黄酮得率的影响,确定三个较为显著的因素(酶解时间、酶量、液料比)为自变量,天名精总黄酮得率为响应值,采用响应曲面统计法中Box-Behnken试验设计方法,建立提取过程中各因素和总黄酮得率的二次多项式模型,并通过响应面优化法分别确定了天名精总黄酮的最佳提取工艺,经修正为:液料比12∶1(mL/g)、酶解时间60 min、酶量0.6%。其中,影响因素按主次顺序排列为:酶量>液料比>酶解时间。优化设计实验条件后,天名精总黄酮的实际得率为7.95%,模型方程理论预测值为7.99%,两者的相对误差为0.50%,验证了实验模型的有效性。由此表明,采用响应面法优化的天名精总黄酮的最佳提取工艺合理可行,可为提取天名精总黄酮提供一定的理论指导和参考依据。To optimize the extraction process of total flavonoids from Carpesium abrotanoides L.Choose enzyme quantity and liquid ratio,ultrasonic time,enzyme digestion temperature and time etc.,respectively,to investigate their effects on day of refined flavonoids extraction rate,determine the three more significant factors(digestion time,enzyme amount,liquid ratio)as the independent variable,the day of refined flavonoids extraction yield as the response value,using response surface method in the Box-Behnken experiment design method.A quadratic polynomial model was established for each factor in the extraction process and the extraction rate of total flavonoids.The optimal extraction conditions were determined by response surface optimization method,which were modified as follows:liquid-solid ratio 12∶1(mL/g),enzymatic hydrolysis time 60 min,and enzyme content 0.6%.Among them,the influencing factors were in the order of enzyme quantity>liquid/solid ratio>enzymolysis time.After optimizing the experimental conditions,the actual extraction rate of total flavonoids was 7.95%,and the theoretical predicted value of the model equation was 7.99%.The relative error of the two was 0.50%,which verified the effectiveness of the experimental model.The results showed that the optimal extraction process of total flavonoids of Carpesium abrotanoides L.optimized by response surface methodology was reasonable and feasible,which could provide theoretical guidance and reference for the extraction of total flavonoids of Carpesium abrotanoides L.
