摘  要:目的:构建鼠源CT-26细胞结肠癌模型评估移植物抗肿瘤(GVT)效应,通过检测磷脂酰丝氨酸(PS)外翻及T淋巴细胞的变化进行判断。方法:通过BALB/c小鼠构建的CT-26结肠癌模型分为A组、B组、C组,其中A组在接种鼠源CT-26结肠癌细胞后不予处理;B组在构建模型后,于第7天输注数量0.5×10^(7)/只脾淋巴细胞、第14天输注数量0.5×10^(7)/只脾淋巴细胞、第21天输注数量1.0×10^(7)/只脾淋巴细胞;C组在小鼠CT-26结肠癌腹腔注射环磷酰胺100 mg/kg(20 mg/mL),同时分别在第7、14、21天输注细胞数量分别为0.5×10^(7)/只、0.5×10^(7)/只、1.0×10^(7)/只脾淋巴细胞。通过流式细胞仪检测外周血CD3^(+)T细胞和腹水中PS外翻表达阳性率。结果:C组CD3^(+)T细胞较A、B组增多(P<0.05);第21天检测PS外翻结果发现,A组PS外翻表达阳性率高于B组(t=2.12,P<0.05)和C组(t=3.74,P<0.05)。结论:T淋巴细胞以及PS外翻可以评估GVT效应抗结肠癌的效果。Objective:To establish a murine CT-26 cell model of colon cancer to evaluate the graft versus tumor(GVT)effect by detecting the changes of phosphatidyl serine(PS)exposure and T lymphocytes.Method:The CT-26 colon cancer model of BALB/c mice was constructed and divided into groups A,B and C.Group A:no treatment after inoculation with CT-26 colon cancer cells;group B:splenic lymphocytes were infused at doses of 0.5×10^(7)/each,0.5×10^(7)/each,1.0×10^(7)/each on days 7,14 and 21 after the CT-26 colon cancer model was constructed;group C:Cyclophosphamide 100 mg/kg(20 mg/mL)intraperitoneally on day 3 after the CT-26 colon cancer model was constructed;group C:cyclophosphamide 100 mg/kg(20 mg/mL)intraperitoneal chemotherapy on day 3 after the construction of mouse CT-26 colon cancer model,and splenic lymphocyte infusion doses of 0.5×10^(7)/each,0.5×10^(7)/each and 1.0×10^(7)/each on days 7,14 and 21,respectively.Flow cytometry was used to detect the peripheral blood CD3T cells and positive rate of PS exposure expression in ascites.Result:The percentage of peripheral blood CD3T cells was increased in group C compared with group A and B(P<0.05);the results of PS exposure detection on day 21 revealed that PS exposure expression was higher in group A than in groups B(t=2.12,P<0.05)and C(t=3.74,P<0.05).Conclusion:PS exposure expression and T lymphocytes can evaluate the GVT effect against colon cancer treatment.
