摘  要:目的 应用网络药理学方法探讨真人养脏汤治疗溃疡性结肠炎的作用机制。方法 利用TCMSP数据库及相关文献筛选真人养脏汤的活性成分及对应靶点,通过GEO、Dis Ge NET、GeneCards、PharmGKB、DrugBank数据库获取与溃疡性结肠炎相关的靶点,并获取药物与疾病的交集靶点,通过STRING数据库和R语言对交集靶点进行蛋白相互作用分析和富集分析,借助Cytoscape3.7.2软件筛选关键靶点和关键成分,然后通过AutoDock软件进行分子对接验证。结果 共筛选出活性成分82个,包括山柰酚、人参皂苷rh2、柯里拉京、甘草查尔酮A、鞣花酸、芒柄花黄素、人参皂苷rf、油酸、玫瑰树碱、肉桂醛等,其作用主要涉及IL6、TNF、IL1B、CASP3、HMOX1、CASP8、CASP1等74个靶点,靶点主要与NF-кB、C型凝集素受体、Toll样受体、NOD样受体、EB病毒感染、人类巨细胞病毒感染、细胞凋亡等信号通路有关;分子对接结果显示关键成分与对应的关键靶点具有较强的结合活性。结论 真人养脏汤可能通过参与炎症、感染、细胞凋亡等相关信号通路及生物功能发挥治疗溃疡性结肠炎的作用。Objective To investigate the mechanism of Zhenren Yangzang Decoction in treating ulcerative colitis based on network pharmacology. Methods Active components of Zhenren Yangzang Decoction and corresponding targets were extracted by TCMSP database and relevant literature. The ulcerative colitis-related targets were screened via GEO, DisGeNET, GeneCards, PharmGKB and DrugBank databases, and the intersection targets of drug and disease were obtained. Protein interaction analysis and enrichment analysis on the overlapping targets were undertaken through STRING database and R language. The key components and targets were screened by Cytoscape 3.7.2 software, and verified by molecular docking using AutoDock softwares. Results A total of 82 active components of Zhenren Yangzang Decoction were screened, including kaempferol, ginsenoside rh2, corilagin, licochalcone a, ellagic acid, formononetin, ginsenoside rf, oleic acid,ellipticine and cinnamaldehyde, and their effects mainly involved 74 targets, including IL6, TNF, ILB, CASP3,HMOX1, CASP8, CASP1, etc. They were mainly related to the signaling pathways, such as NF-κB, C-type lectin receptor, Toll-like receptor, NOD-like receptor, Epstein-Barr virus infection, human cytomegalovirus infection, apoptosis, etc. The results of molecular docking showed that the key componets had strong binding activity with corresponding targets. Conclusion Zhenren Yangzang Decoction may play a role in treating ulcerative colitis by participating in inflammation, infection, apoptosis-related signaling pathways and biological functions.
