摘 要:以尖果沙枣种子为研究对象,对沙枣种子总黄酮的提取条件和体外抗氧化活性进行了初步研究。以提取次数、料液比、提取时间、乙醇体积分数作为考查因素,设计正交试验优化提取工艺。以芦丁为标准品,采用硝酸铝-亚硝酸钠比色法,用紫外可见分光光度法测定总黄酮的含量。结果表明,沙枣种子总黄酮的最佳提取工艺为乙醇体积分数70%,提取次数3次,料液比1∶30,提取时间0.5 h。在此条件下,沙枣种子总黄酮含量最高,为5.102 mg/g。体外抗氧化试验结果表明,沙枣种子总黄酮具有体外清除DPPH自由基的作用。研究确定了超声波提取沙枣种子总黄酮的最佳工艺参数,证明沙枣种子总黄酮具有一定的体外抗氧化活性,为沙枣黄酮类化合物在天然药物领域的进一步开发和研究提供参考。The extraction conditions and antioxidant activity of total flavonoids from Elaeagnus oxycarpa Schlechtend seeds were studied.Taking extraction times,solid-liquid ratio,extraction time and ethanol volume fraction as factors,orthogonal experiment was designed to optimize the extraction process.Rutin was used as standard,and the content of total flavonoids was determined by visible-ultraviolet spectrophotometer with aluminum nitrate-sodium nitrite colorimetry.The results showed that the optimum extraction process of total flavonoids from Elaeagnus oxycarpa Schlechtend seeds were as follows:ethanol concentration 70%,extraction 3 times,solid-liquid ratio 1∶30,extraction time 0.5 h.Under these conditions,the consent of flavonoids from Elaeagnus oxycarpa Schlechtend seeds was the highest,with flavonoid content was 5.102 mg/g.The results of antioxidant activity in vitro showed that total flavonoids from Elaeagnus oxycarpa Schlechtend seeds had DPPH free radical scavenging effect in vitro.The optimum technological parameters of ultrasonic extraction of total flavonoids from Elaeagnus oxycarpa Schlechtend seeds were determined,which proved that flavonoids from Elaeagnus oxycarpa Schlechtend seeds had certain antioxidant activity in vitro,and provided reference for further development of flavonoids from Elaeagnus oxycarpa Schlechtend in natural medicine.
