摘 要:目的:优化穿山龙总皂苷的提取工艺并测定其体外抗氧化性。方法:以总皂苷的得率为评价指标,基于单因素实验结合响应面法优化总皂苷的提取工艺参数,并在最优工艺条件下提取总皂苷,采用大孔吸附树脂对其进行分离纯化,对纯化前后的总皂苷进行体外抗氧化活性研究。结果:在乙醇浓度为80%、提取时间为80 min、提取温度为80℃、超声时间为25 min条件下,提取物中穿山龙总皂苷的含量为16.24%。体外抗氧化实验研究表明,穿山龙总皂苷具有较强的抗氧化性,未纯化的总皂苷对DPPH、ABTS清除率的IC50值分别为4.9、2.8 mg/mL,纯化后的总皂苷对DPPH、ABTS清除率的IC50值分别是3.8、2.2 mg/mL。结论:采用响应面方法优化的穿山龙总皂苷提取工艺有效可靠,稳定可行;提取的穿山龙总皂苷具有很强的抗氧化活性,可作为天然抗氧化剂。Objective:To optimize the extraction process of total saponins of Rhizoma Dioscorea Nipponicae and determine its in vitro antioxidant activity. Methods:The total saponins was taken as an evaluation index,the extraction process parameters of total saponins was optimized based on single-factor experiment combined with response surface method. The total saponins before and after purification were studied for in vitro antioxidant activity. Results:It was found that when the ethanol concentration was 80%,the extraction time was 80 minutes,the extraction temperature was 80 ℃,and the ultrasonic time was 25 minutes,the content of the total saponin was 16. 24%. Anti-oxidant experiments in vitro showed that the total saponins of Rhizoma Dioscorea Nipponicae had strong anti-oxidant properties. The IC50values of the unpurified saponins for the clearance rates of DPPH and ABTS were 4. 9 mg/mL and 2. 8 mg/mL,respectively;the IC50values of the purified saponins for the clearance rates of DPPH and ABTS were 3. 8 mg/mL and 2. 2 mg/mL,respectively.Conclusion:The extraction process of Rhizoma Dioscorea Nipponicae optimized by single-factor response surface method is effective,reliable,stable and feasible. The total saponins of Rhizoma Dioscorea Nipponicae has a strong anti-oxidant activity,which can be used as a natural anti-oxidant agent.
