复方黄杞颗粒对实验性自身免疫性重症肌无力大鼠模型IL-6、IL-17、SI、AchRAb影响

摘　　要：目的观察复方黄杞颗粒对自身免疫性重症肌无力大鼠模型(EAMG)血清白介素-6(IL-6)、白介素-17(IL-17)、铁(SI)、乙酰胆碱受体抗体(AchRAb)的调节作用。方法选择雌性Lewis大鼠60只,随机分为空白对照组、模型对照组、实验药物组、溴吡斯的明组。采用二次免疫注射法建立实验性自身免疫重症肌无力大鼠模型。二次免疫1周后,空白对照组给予5.4 mg/(kg·d)生理盐水灌胃,1次/d,持续21 d;模型对照组给予5.4 mg/(kg·d)生理盐水灌胃,1次/d,持续21 d;溴吡斯的明组给予5.4 mg/(kg·d)溴吡斯的明灌胃给药,1次/d,持续21 d;实验药物组给予5.4 mg/(kg·d)复方黄杞颗粒灌胃给药,1次/d,持续21 d。比较各组大鼠血清IL-6、IL-17、SI、AchRAb的浓度。结果模型对照组血清中AchRAb、IL-6、IL-17浓度明显高于空白对照组(P<0.05)。实验药物组、溴吡斯的明组血清中AchRAb、IL-6、IL-17浓度明显低于模型对照组(P<0.05)。各组大鼠SI浓度与模型对照组比较,差异无统计学意义(P>0.05)。结论复方黄杞颗粒可影响自身免疫性重症肌无力大鼠模型血清AchRAb、IL-6、IL-17水平。Objective To observe the regulation of Compound Huangqi Granule(复方黄杞颗粒)on serum IL-6,IL-17,SI,AchRAb in autoimmune myasthenia gravis rat model.Methods Sixty female Lewis rats were randomly divided into blank control group,model control group,experimental drug group and bromopyrimidine group.The experimental autoimmune myasthenia gravis rat model was established by secondary immune injection.One week after the second immunization,the blank control group was given 5.4 mg/(kg·d)normal saline by gavage,once a day for 21 days;The model control group was given 5.4 mg/(kg·d)normal saline by gavage once a day for 21 days;In the bromopyrimidine group,5.4 mg/(kg·d)bromopyrimidine was administered by gavage once a day for 21 days;The experimental drug group was given Compound Huangqi Granule by gavage at the rate of 5.4 mg/(kg·d)once a day for 21 days.The serum concentrations of IL-6,IL-17,SI and AchRAb in the blank control group,model control group,experimental drug group and bromadigmine group were compared.Results The serum concentrations of AchRAb,IL-6 and IL-17 in the model control group were significantly higher than those in the blank control group(P<0.05).The concentrations of AchRAb,IL-6 and IL-17 in serum of myasthenia gravis model rats in the experimental drug group and bromopyrimethamine group were significantly lower than those in the model control group(P<0.05).There was no significant difference in SI concentration between rats in each group and model control group(P>0.05).Conclusion Compound Huangqi Granule can affect the levels of serum AchRAb,IL-6 and IL-17 in EAMG.
