摘 要:为研究不同分子质量水溶性大豆皮膳食纤维对糖尿病模型小鼠糖脂代谢及血糖耐量的影响,分别给予四氧嘧啶致糖尿病模型小鼠低(2.50 g/kgbw)、中(3.75 g/kgbw)、高(5.00 g/kgbw)剂量的不同分子质量3种水溶性大豆皮膳食纤维(SDF-1、0.70~2.96 ku;SDF-2、2.95~31.03 ku和SDF-3、36.53~338.10 ku),以血糖、体重、血清总胆固醇(TC)、血清甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)、血清胰岛素含量(FIN)、胰岛素敏感指数(ISI)、胰岛素抵抗指数(HOMA-IR)、肝脏指数和肾脏指数、超氧化物歧化酶(T-SOD)活性、血糖曲线下面积等为指标,进行糖脂代谢试验,及低剂量血糖耐量试验。试验结果显示:较糖尿病模型对照组,所有剂量组小鼠的血糖降低、体重上升、肝脏指数和肾脏指数的降低、血糖曲线下面积的减小、TC和TG的降低均具有非常显著的差异(P<0.01);除SDF-2低剂量组外,其他剂量组FIN值升高均具非常显著的差异(P<0.01);仅SDF-1中剂量组的HOMA-IR降低、SDF-1和SDF-2的中高剂量组的T-SOD升高具有显著的差异(P<0.05);而ISI的恢复,及LDL-C的降低,不具显著性。说明不同分子质量、不同剂量的3种水溶性大豆皮膳食纤维对糖尿病模型小鼠糖脂代谢异常具有缓解作用,并增强对葡萄糖的耐受能力,且因分子质量大小、单糖的组成、吸水及膨胀性、吸油、吸附羟自由基和胆固醇的能力以及黏性等性质不同而存在效果的差异,总体效果SDF-3>SDF-1>SDF-2,可能与水溶性大豆皮膳食纤维杂多糖中存在果糖聚合的支链或片段有关,且单糖的组成中果糖含量高、黏性大、吸水及吸水膨胀性强、对羟自由基的清除能力、对胆固醇吸附能力强,有利于对糖尿病模型小鼠糖脂代谢异常的缓解及糖耐量的增强。To explore the effects of soluble dietary fiber with different molecular masses from soybean hulls on glucose and lipids metabolism and blood glucose tolerance of diabetic mice.Alloxan-induced diabetes model mice were studied for glucose and lipids metabolism and low-dose were designed for blood glucose tolerance test.Intake dose was designed to low(2.50 g/kgbw),medium(3.75 g/kgbw)and high(5.00 g/kgbw)doses of three soluble dietary fibers with different molecular mass(SDF-1,0.70~2.96 ku;SDF-2,2.95~31.03 ku and SDF-3,36.53~338.10 ku)from soybean hulls.Blood sugar,body weight,serum total cholesterol(TC),serum triglycerides(TG),low density lipoprotein(LDL-C),high density lipoprotein(HDL-C),serum insulin content(FIN),insulin sensitivity index(ISI),insulin eesistance index(HOMA-IR),liver index and kidney index,superoxide dismutase activity(T-SOD)and area under the blood sugar curve,etc.,were used as the indicators.Experimental results:compared with the diabetic model control group,all doses significantly increased body weight,reduced blood sugar,liver index and kidney index,area under the blood sugar curve,lower TC and TG of mice(P<0.01).Except for the SDF-2 low-dose group,the increase in FIN value of the other dose groups had very significant differences(P<0.01).Only HOMA-IR of the SDF-1 medium-dose group decreased,and T-SOD of the SDF-1 and SDF-2 high-dose groups increased significantly(P<0.05);however,the recovery of ISI and the reduction of LDL-C were not significant.Three kinds of soluble dietary fiber with different molecular masses and dosages from soybean hulls could alleviate the abnormal glucose and lipids metabolism,enhanced glucose tolerance in diabetic mice.Due to the different molecular masses,monosaccharide compositions and properties,such as water absorption and swelling,oil absorption,hydroxyl radical and cholesterol adsorption ability,and viscosity,etc.,the overall effect was SDF-3>SDF-1>SDF-2.Because the soluble dietary fiber from soybean hulls,a heteropolysaccharide,might be branched chains or frag
