摘　　要：目的:探索碳酸酐酶9(CA9)在舌鳞状细胞癌(TSCC)中的表达及预后意义。方法:从癌症基因组图谱(TCGA)数据库下载舌鳞癌中CA9的RNA-Seq数据及临床资料。分析癌及癌旁组织中CA9的表达差异,及其与临床病理参数及预后的关系。基因集富集分析(GSEA)方法探讨CA9在舌鳞癌中潜在功能。结果:获得147例TSCC组织,15例癌旁组织和156例TSCC患者信息。CA9在TSCC组织中高表达,其表达水平与T分期相关(P=0.007),CA9高表达患者死亡率高于低表达患者(P=0.001)。CA9是TSCC患者的独立预后因素(P<0.001)。GSEA结果表明CA9在糖酵解与糖异生调节、细胞周期调控、同源重组等通路高度富集。结论:CA9是舌鳞癌的独立预后因素,可能成为舌鳞癌的潜在预后标志物和治疗靶点。Objective: To investigate the prognostic significance and the expression of carbonic anhydrase 9(CA9) in tongue squamous cell carcinoma(TSCC) tissues. Methods: Data of TSCC with the CA9 gene expression profile and clinical information were downloaded from the cancer genome atlas(TCGA) database.CA9 expression in cancer and corresponding adjacent tissues, the correlation between CA9 gene expression and clinicopathological features and prognosis of TSCC patients were analyzed. Gene set enrichment analysis(GSEA) method was used to predict the potential molecular function of CA9 in TSCC. Results: The mRNA level of CA9 in TSCC tissues was higher than that in corresponding adjacent tissues. CA9 expression level was correlated with T stage(P=0.007). The mortality rate of patients with high CA9 expression was higher than that of patients with low CA9 expression(P=0.001). CA9 was an independent prognostic factor for TSCC prognosis(P<0.001). GSEA analysis showed that CA9 might be involved in the development of TSCC by regulating glycolysis, gluconeogenesis, cell cycle and homologous recombination. Conclusion: CA9 is an indepen-dent prognostic factor for TSCC and might be a potential prognostic biomarker and therapeutic target for TSCC.
