摘  要:目的:分析5岁孤独症谱系障碍患儿唾液菌群结构。方法:纳入30例健康(H组)和30例孤独症谱系障碍(ASD组)5岁儿童,共采集60份唾液样本,运用SMRT测序分析细菌群落结构,分析疾病状态对唾液细菌代谢的影响,建立疾病风险预测模型。结果:ASD组群落丰富度减少,微生物菌群落更保守(P<0.05),鉴别出ASD组高丰度细菌(P<0.05),如嗜血杆菌和奈瑟菌,两组菌群在营养物质代谢方面存在差异(P<0.05)。基于唾液链球菌、血链球菌和未命名的奈氏菌3种细胞在种水平差异(LDA>4)建立的孤独症谱系障碍风险预测模型区分健康和疾病状态的准确率达80.6%以上。结论:特定的唾液微生物群有助于评估和筛选孤独症谱系障碍风险。Objective: To study the salivary microflora structure of 5-year-old children with autism spectrum disorder(ASD). Me-thods: Salivary microbiome of the children aged 5-year-old were collected from 2 groups: ASD group(n=30) and healthy developing group(H;n=30). The full length of 16 S rRNA gene was analyzed by PacBio SMRT technology, and the bacterial community structure was analyzed to predict the influence of disease status on the metabolism of saliva bacteria, and the risk assessment model of ASD was established. Results: The species diversity and richness of the salivary microflora of ASD group were lower than those of the H group(P<0.01). The community structure of ASD group was more conserved(P<0.05). High abundance of microorganisms in group ASD was distinguished(P<0.05), such as Neisseria and Haemophilus. Metabolic function of the bacteria between the ASD and H groups was significantly different(P<0.05). Eventually, 3 sepcies ratio of microbial, Streptococus salivarius, streptococcus sanguinis and unnamed Neisseria, distinguished ASD from healthy subjects(80.6% accuracy). Conclusion: Specific salivary microbiotia may be useful in assessing and screening for ASD risk.
