摘 要:目的通过分析俞氏安肠方(YST)对UC小鼠炎症因子、NLRP3炎症小体及结肠黏膜屏障的影响,来探讨其治疗UC的潜在机制。方法实验小鼠随机分成对照组、模型组、YST低剂量组、高剂量组,用2,4,6-三硝基苯磺酸(TNBS)构建UC小鼠模型。记录所有小鼠的体质量、粪便黏度和粪便出血情况。处死后,用苏木精-伊红染色观察结肠组织病理变化。ELISA检测结肠组织TNF-α、IL-1β、IL-6的水平。Western Blot分析NLRP3炎症小体和结肠黏膜屏障相关的蛋白质的表达。结果TNBS导致实验小鼠DAI值升高,引发严重的组织学损伤。YST可以降低小鼠结肠组织的TNF-α、IL-1β、IL-6水平。YST可以下调NLRP3炎症小体相关的蛋白(NLRP3、ASC、Caspase-1)表达水平。YST可以上调结肠黏膜屏障相关蛋白(ZO-1、Occludin、Claudin-1)的表达。结论YST可以通过减少炎症反应、抑制NLRP3炎症小体和保护肠黏膜屏障来改善TNBS诱导的UC小鼠的症状及组织学损伤。Objective By analyzing the effects of YST on inflammatory factors,NLRP3 inflammasome and colonic mucosal barrier in UC mice,the potential mechanism of YST in treating UC was explored.Methods The experimental mice were randomly divided into control group,model group,YST low dose group and high dose group,and UC mouse model was constructed with 2,4,6-trinitrobenzene sulfonic acid(TNBS).Weight,fecal viscosity,and fecal bleeding were all recorded in all mice.After death,the pathological changes of colon tissue were observed with Sumu-Ihhong dyeing.The levels of colon tissue TNF-α,IL-1β,and IL-6 were detected by ELISA.Western Blot was adopted to analyze the expression of proteins associated with NLRP3 inflammatory small bodies and the colonic mucosal barrier.Results TNBS led to an increase in DAI in experimental mice,causing severe histological damage.YST can reduce TNF-α,IL-1β,IL-6 levels in colon tissue in mice.YST can lower the expression levels of NLRP3 inflammatory small body-related proteins(NLRP3,ASC,Caspase-1).YST can increase the expression of coliform mucous membrane barrier-related proteins(ZO-1,Occludin,Claudin-1).Conclusion YST can improve the symptoms and histological damage of TNBS-induced UC mice by reducing inflammatory response,suppressing NLRP3 inflammatory small bodies and protecting the intestinal mucosa barrier.
