摘 要:目的构建黄芩苷纳米水凝胶并评估其对实验性牙周炎大鼠的影响。方法采用乳化-蒸发法制备壳聚糖/二油酰基卵磷脂-黄芩苷纳米水凝胶,用动态光散射、扫描电子显微镜和光谱仪法测定其性质。选取30只5周龄雄性Wistar大鼠随机分为5组(n=6),即正常组、模型组(牙周炎对照组)、对照组(空白纳米水凝胶)、阳性对照组(洗必泰)和实验组(黄芩苷纳米水凝胶);与上述治疗第2周和第4周检测牙周探查深度、具核梭杆菌和牙龈卟啉单胞菌数量;然后处死各组大鼠,通过组织病理学对组织进行评估并检测牙周组织中IL-1β、IL-6、IL-8和TNF-α水平变化。结果黄芩苷纳米水凝胶具有良好的透气性,粒径为577.0nm,分散指数为0.81,表面ZETA电位为46.30mV,包封率为(98.56±0.23)%,载药率最高为(14.21±0.25)%,体外释放性能良好;动物实验表面能显著改善牙周袋的深度和牙周致病菌的数量(P<0.05),在牙周组织中检测到IL-1β、IL-6、IL-8和TNF-α的表达降低。结论黄芩苷纳米水凝胶具有良好的缓释作用,黄芩苷缓释组分能减轻牙周炎症,抑制牙周致病微生物,在牙周炎治疗中具有良好的应用前景。Objective To construct the Baicalin Nanohydrogel and to evaluate its effect on experimental periodontitis in rats. Methods Chitosan/dioleoyl lecithin-baicalin nanohydrogels were prepared by emulsification-evaporation method, and their properties were determined by dynamic light scattering, scanning electron microscope and spectrograph.Thirty five-week-old male Wistar rats were randomly divided into 5 groups(n=6), normal group, model group(periodontitis control group), control group(blank nano-hydrogel), positive control group(chlorhexidine) and experimental group(baicalin nano-hydrogel). The depth of periodontal exploration, the numbers of FN and P. Gingivalis were measured at the 2 nd and 4 th week of treatment, and then the rats were killed, the changes of IL-1β, IL-6, IL-8 and TNF-α in periodontal tissues were evaluated by histopathology. Results Baicalin Nanoparticle Hydrogel had good permeability. The particle size was 577.0 nm, dispersion index was 0.81, surface ZETA potential was 46.30 mV, entrapment efficiency was(98.56±0.23)%, drug loading rate was(14.21±0.25)%. The depth of periodontal pockets and the number of periodontal pathogens were significantly improved(P<0.05),and the expression of IL-1β, IL-6, IL-8 and TNF-α were decreased in periodontal tissues. Conclusion Baicalin loaded nano-microspheres Hydrogel has good sustained release effect. Baicalin sustained release components can reduce periodontal inflammation, inhibit periodontal pathogenic microorganisms, has a good application prospect in the treatment of periodontitis.
