摘  要:目的 探讨细胞周期蛋白B1(CCNB1)和自噬标记蛋白Beclin1在宫颈癌组织中的表达及意义。方法 选取2018年1月—2021年6月在台州市第一人民医院妇科住院的60例宫颈癌患者的标本,采用实时荧光定量PCR(RTPCR)法检测60例宫颈癌组织及60例癌旁组织CCNB1 mRNA和Beclin1 mRNA,采用免疫组织化学(SP)法检测宫颈癌组织及癌旁组织中的CCNB1和Beclin1,用SPSS 22.0软件对结果进行统计学分析。结果 与癌旁组织相比较,宫颈癌组织中CCNB1 mRNA表达水平升高(P<0.05),Beclin1 mRNA表达水平降低(P<0.05)。宫颈癌组织CCNB1阳性率高于癌旁组织(P<0.05),Beclin1阳性率低于癌旁组织(P<0.05)。分化程度越低、有淋巴结转移、TNM分期越高、浸润深度越深的CCNB1表达越高(P<0.05),Beclin1表达越低(P<0.05)。结论 CCNB1在宫颈癌组织中表达上调,Beclin1在宫颈癌组织中表达下调,与宫颈癌的发生发展有关。Objective To explore the expression and significance of Cyclin B1( CCNB1) and Beclin1 in cervical cancer tissues.Methods The specimens of 60 patients with cervical cancer who were hospitalized in the Department of Gynecology,Taizhou First People.s Hospital from January 2018 to June 2021 were selected.The real-time fluorescent quantitative PCR(RT-PCR) method was used to detect 60 cases of cervical cancer tissue and 60 cases of adjacent cancer.Immunohistochemistry(SP) was used to detect CCNB1 and Beclin1 in cervical cancer tissues and adjacent tissues,and the results were statistically analyzed with SPSS 22.0 software.Results The level of CCNB1mRNA in cervical cancer tissues was significantly higher than that in adjacent tissues(P< 0.05),and the level of Beclin1 mRNA in cervical cancer tissues was significantly lower than that in adjacent tissues(P< 0.05).The positive rate of CCNB1 in cervical cancer tissues was significantly higher than that in adjacent tissues(P< 0.05),and the positive rate of Beclin1 in cervical cancer tissues was significantly lower than that in adjacent tissues(P< 0.05).The lower the degree of differentiation,lymph node metastasis,the higher the TNM stage,and the deeper the depth of invasion,the higher the expression of CCNB1(P< 0.05) and the lower the expression of Beclin1(P< 0.05).Conclusion CCNB1 is up-regulated in cervical cancer tissues,Beclin1 is down-regulated in cervical cancer tissues,which is related to the occurrence and development of cervical cancer.
