摘 要:旨在探究单增李斯特菌(Listeria monocytogenes,Lm)溶血素hly、llsB基因对小鼠肠道的影响。在构建Lm的hly基因缺失株基础上,以Lm90SB2及溶血素基因缺失株Lm90SB2-Δhly、Lm90SB2-ΔllsB为试验菌,进行生长曲线、溶血活性以及器官载菌量的测定。结果:缺失株在不同条件下的生长特性与亲本株相比无明显差异;Lm90SB2、Lm90SB2-ΔllsB的溶血效价分别为1log2、2log2,而Lm90SB2-Δhly基本无溶血现象。缺失株Lm90SB2-Δhly、Lm90SB2-ΔllsB的肠道载菌量均低于亲本株Lm90SB2(P<0.05),在各个时间点肠道中的细菌定殖率依次为Lm90SB2>Lm90SB2-Δhly>Lm90SB2-ΔllsB。本研究表明,hly、llsB基因对Lm在小鼠肠道内定殖可能有直接或间接的调控作用,试验结果为进一步研究hly、llsB基因在Lm致病过程中的作用提供一定的理论依据。This study was to determine the effects of the hemolysin hly,llsB genes deletion in Listeria monocytogenes on the intestinal tract of mice. The Listeria monocytogenes Lm90 SB2 and the hemolysin gene deletion strains Lm90 SB2-Δhly and Lm90 SB2-ΔllsB were used as the tested strains. The growth curves, hemolysis activity and intestinal bacterial load were measured in the mice. The results showed that there was no significant difference in the growth characteristics of the deletion strain, as compared with its parents. The hemolysis tits of Lm90 SB2 and Lm90 SB2-ΔllsB were 1 log2 and 2 log2, respectively, while Lm90 SB2-Δhly showed no hemolysis. The intestinal bacterial loads of the deletion strains Lm90 SB2-Δhly and Lm90 SB2-ΔllsB were both lower than that of the parent strain Lm90 SB2(P<0.05), and their intestinal bacterial colonization rates at each time point of the test were in the order of Lm90 SB2>Lm90 SB2-Δhly>Lm90 SB2-ΔllsB. However, their bacterial colonization rates in the brain and spleen of the mice was in the order of Lm90 SB2>Lm90 SB2-ΔllsB>Lm90 SB2-Δhly. In conclusion, the hly and llsB genes might have directly or indirectly regulated the intestinal tract of the mice. These results provided a theoretical basis for further research on the pathogenesis of the hly and llsB genes and on prevention and control of listeriosis.
