摘 要:目的 探讨姜黄素干预肾上腺皮质癌SW-13细胞的miRNA调控机制。方法 将肾上腺皮质癌SW-13细胞分为对照组和姜黄素干预组,分别加入基础培养基2 mL、50μmol/L姜黄素溶液2 mL,干预24 h后提取总RNA用于小RNA文库构建和测序。将获得的序列进行miRNA注释和新miRNA预测后,筛选差异性表达的miRNA;对差异性表达的miRNA进行靶基因预测,对靶基因进行功能和通路富集分析。使用实时荧光定量PCR鉴定部分差异性表达miRNA的表达量。结果 共筛选出44个差异性表达的miRNA,包括21个表达上调的miRNA和23个表达下调的miRNA,其中预测有靶基因且有功能的miRNA共9个。功能富集分析结果显示,靶基因主要富集在细胞增殖、细胞迁移、细胞分化和凋亡等生物学过程;通路富集分析结果显示,靶基因主要富集在癌症途径、丝裂原活化蛋白激酶信号通路、内质网应激信号通路、细胞周期和凋亡途径等。实时荧光定量PCR结果提示,miR-184、miR-490-3p、miR-766-3p、miR-200b-3p、miR-200c-3p和miR-3622a-5p的表达量与小RNA测序数据结果基本一致。结论 姜黄素可能通过miR-184、miR-490-3p、miR-766-3p、miR-200c-3p、miR-3622a-5p等miRNA参与调控癌症途径、丝裂原活化蛋白激酶信号通路、内质网应激信号通路、细胞周期和凋亡途径等信号通路,以抑制细胞增殖、迁移、分化和凋亡,从而发挥抗肾上腺皮质癌的作用。Objective To explore the miRNA regulatory mechanism of Curcumin in the intervention of adrenocortical carcinoma SW-13 cells.Methods Adrenocortical carcinoma SW-13 cells were assigned to control group or Curcumin intervention group added with 2 mL basal culture medium and 2 mL Curcumin solution with the concentration of 50 μmol/L,respectively.After 24 hours of intervention,the total RNA was extracted for small RNA library establishment and sequencing.After miRNA annotation and new miRNA prediction of the obtained sequences,differentially expressed miRNAs were screened;furthermore,the target gene prediction was conducted in differentially expressed miRNAs,and the functional and pathway enrichment analysis of the target genes was conducted.The expressions of some differentially expressed miRNAs were verified by the real-time fluorescence quantitative PCR.Results A total of 44 differentially expressed miRNAs were screened,including 21 miRNAs of up-regulated expression,and 23 miRNAs of down-regulated expression,thereinto a total of 9 miRNAs were predicted to possess target genes and function.The results of the functional enrichment analysis revealed that the target genes were mainly enriched in the biological procedures with respect to cell proliferation,cell migration,cell differentiation and apoptosis,etc.The results of the pathway enrichment analysis depicted that the target genes were mainly enriched in cancer approach,mitogen-activated protein kinase signaling pathway,endoplasmic reticulum stress signaling pathway,cell cycle and apoptosis approach,etc.The results of the real-time fluorescence quantitative PCR exhibited that the expressions of miR-184,miR-490-3 p,miR-766-3 p,miR-200 b-3 p,miR-200 c-3 p,and miR-3622 a-5 p were basically consistent with the results of small RNA sequencing data.Conclusion Curcumin may be involved in regulating signaling pathways in terms of cancer approach,mitogen-activated protein kinase signaling pathway,endoplasmic reticulum stress signaling pathway,cell cycle and apoptosis appro
