摘 要:目的:观察柴胡皂苷d(saikosaponins-d,SSd)对肝星状细胞TGFβ_(1)、Smad3/7mRNA表达的影响。方法:采用大鼠肝星状细胞(hepatic stellate cell-T6,HSC-T6)为模型,采用药物进行干预后加入H_(2)O_(2)诱导氧化应激,采用RT-PCR实验方法检测TGFβ_(1)、Smad3/7 mRNA表达。结果:与空白组比较,模型组TGFβ_(1)、Smad3表达增强,Smad7表达减弱(P<0.05)。与模型组比较,SSd组干预后HSC-T6细胞中TGFβ_(1)、Smad3 mRNA水平降低,Smad7 mRNA水平增高(P<0.05)。这种作用可被雌激素受体(estrogen receptor,ER)完全拮抗剂和ERβ拮抗剂阻滞。结论:SSd能下凋TGFβ_(1)、Smad3 mRNA表达,上调Smad7 mRNA的表达,对HSC-T6的TGFβ-Smad信号通路有明显的影响,可能是SSd对肝纤维化产生治疗作用的机制之一。Objective:To observe the effects of SSd on the expressions of TGFβ_(1),Smad3/7mRNA in hepatic stellate cells.Methods:Rat HSC-T6 were adopted as the model,after drug intervention,H_(2)O_(2) was added to induce oxidative stress,RT-PCR was used to detect the expressions of TGFβ_(1),Smad3/7mRNA.Results:Compared with the blank group,the expression of TGFβ_(1)and Smad3 in the model group was enhanced,while the expression of Smad7 was weakened(P<0.05).Compared with the model group,the levels of TGFβ_(1)and Smad3 mRNA were decreased while the level of Smad7mRNA was elevated in HSC-T6 cells in SSd group after the intervention(P<0.05).The effects could be blocked by ER complete antagonist and ERβantagonist.Conclusion:SSd could down regulate the expressions of TGFβ_(1),Smad3 mRNA,up regulate the expressions of Smad7 mRNA,and it could affect TGFβ-Smad signal pathway in HSC-T6 signal pathway,which might be one of the mechanisms of SSd producing therapeutic effects on liver fibrosis.
