摘 要:目的基于数据挖掘,研究CLEC16A基因与混合性肾癌(KIPAN)发展、预后的关系及意义。方法从TCGA数据库中下载KIPAN的CLEC16A基因表达谱及其临床信息,利用Perl脚本和R包进行处理并分析之间的关系。结果CLEC16A在KIPAN中的表达较正常组织低,且随着KIPAN临床分级的提高,CLEC16A的表达呈下调趋势。CLEC16A低表达与KIPAN预后不良呈正相关(总生存率:HR=0.69,P=0.006;无病生存率:HR=0.72,P=0.028)。基因富集分析结果显示,CLEC16A高表达主要在溶酶体和氧化磷酸化等生物学过程或通路存在富集。而CLEC16A低表达在抗原加工及呈递、免疫球蛋白A(Ig A)肠道免疫网络产生、白细胞跨内皮迁移、NOD样受体信号通路、自然杀伤细胞介导的细胞毒性等信号通路中存在富集。结论CLEC16A可能作为KIPAN发生的抑制物,其低表达是肾癌进展及不良预后的危险因素,高表达可能是抑制KIPAN发生的保护因子。Objective To study the relationship and significance of CLEC16A gene and the development and prognosis of mixed renal carcinoma(KIPAN)based on data mining.Methods The CLEC16A gene expression profile of KIPAN and its clinical information were downloaded from the TCGA database.The Perl script and R package were used to process the data and analyze the relationship.Results The expression of CLEC16A in KIPAN was lower than that in normal tissues,and with the improvement of KIPAN clinical grade,the expression of CLEC16A showed a downward trend.Low CLEC16A expression was positively correlated with poor prognosis in KIPAN(overall survival HR=0.69,P=0.006;disease-free survival:HR=0.72,P=0.028).The results of gene set enrichment analysis showed that the high expression of CLEC16A was mainly enriched in biological processes or pathways such as lysosome and oxidative phosphorylation.The low expression of CLEC16A was enriched in signaling pathways such as antigen processing and presentation,production of IGA intestinal immune network,leukocyte trans endothelial migration,NOD-like receptor signaling pathway,and natural killer cell-mediated cytotoxicity.Conclusion CLEC16A may act as an inhibitor of the occurrence of KIPAN,its low expression is a risk factor for the progression and poor prognosis of renal cancer,and its high expression may be a protective factor against the occurrence of KIPAN.
