摘  要:目的检测分析长链非编码RNA-BANCR在胰腺癌中的表达情况及临床意义,并探究其对胰腺癌细胞增殖、迁移、侵袭和淋巴管生成能力的影响。方法实时定量PCR方法检测23例2016年1月至2020年12月手术切除的同一患者胰腺癌以及瘤旁组织中BANCR表达水平,分析其与胰腺癌临床病理特征及预后的相关性。实时定量PCR方法检测BANCR在胰腺癌细胞系中表达情况,在PANC-1细胞中转染BANCR沉默表达质粒后通过细胞增殖实验(CCK8)、细胞迁移实验(Transwell)、划痕实验检测和体外淋巴管内皮细胞成管实验检测BANCR对胰腺癌细胞增殖、迁移能力以及微淋巴管生成能力的影响。实验数据应用SPSS23.0统计学软件进行统计分析。BANCR的表达水平及微淋巴管生成密度(MLVD)以(x±s)表示,组间对比采用独立样本1检验。以P<0.05为差异有统计学意义。结果BANCR在胰腺瘤中的表达水平(2.764±0.358)高于癌旁胰腺组织(1.299±0.192),且在细胞学实验中也证实BANCR在胰腺癌细胞系中表达水平升高。结合临床病理特征,淋巴结转移阳性组中BANCR的表达水平(3.924±1.781)显著高于淋巴结转移阴性组(1.257±0.360),差异有统计学意义(P<0.001)。BANCR高表达组患者预后较差。转染siRNA干扰BANCR表达后胰腺癌细胞增殖、迁移、迁移能力显著下降,体外淋巴管内皮细胞成管能力(10.000±1.581)也较对照组(转染si-NC组)(13.400±2.074)降低。结论BANCR在胰腺癌组织中高表达,且与胰腺癌淋巴结转移、预后差相关。胰腺癌细胞中,BANCR具有促进细胞增殖、迁移、侵袭以及微淋巴管生成的作用,有望为胰腺癌的诊治提供新靶点。Objective To investigate the expresion and clinical relevance of LncRNA BANCR in pancreatic cancer tissues,and to explore the effect of BANCR on the proliferation,migration,invasion and lymphangiogenesis of pancreatic cancer cells.Methods The expression level of BANCR in pancreatic cancer and adjacent tissues of 23 patients surgically resected from January 2016 to December 2020 was detected by real-time quantitative PCR,and its correlation with clinicopathological features and prognosis of pancreatic cancer was analyzed.Real-time quantitative PCR was used to delect the expression of BANCR in pancreatic cancer cellines.The effects of BANCR on the proliferation,migration and lymphangiogenesis of pancreatic cancer cells were detected by cell proliferation assay(CCK8),cll migration assay(Transwell),scratch assay and lymphatic endothelial ell tube fomation assay in vitro.The experimental data were analyzed by SPSS 23.0 statistical software.The expression level of BANCR and the microlymphangiogenic density(MLVD)were expresed as(x±s),and the comparison between groups was analyzed by independent sample t test.P<0.0S was considered statisticaly signifiant.Results The expression level of BANCR in pancreatic cancer(2.764±0.358)was higher than that in adjacent pancreatic tissues(1.299±0.192),and the expression level of BANCR in pancreatic cancer cell lines was also confirmed by cytological experiments.Combined with clinicopathological features,the expression level of BANCR in lymph node metastasis positive group(3.924±1.781)was significantly higher than that in lymph node metastasis negative group(1.257±0.360),and the diference was satisially significant(P<0.001).Patients with high BANCR expresion had poor prognosis.The prlfeation,migation and migration abilty of pancreatic cancer cells were significantly decreased after siRNA transfection with BANCR expression,and the tube fomation ability of lymphatic endothelial cels in vitro(10.000±1.581)was also decreased compared with the control group(13.400±2.074).Conclusion B
