摘  要:通过水提醇沉法对柠檬桉树脂的多糖(ERP)进行提取,采用苯酚-硫酸法测定多糖含量,得到柠檬桉树脂粗多糖(ERPC)含量为7.79%。通过Sevage法和三氯乙酸法(TCA)对ERP进行脱蛋白,结果表明TCA法对ERP脱蛋白效果更好,且在TCA浓度达到6%时,蛋白清除率达到94.79%,多糖的保留率为82.99%。采用聚酰胺层析柱对ERP进行除色素,脱色率为77.95%,多糖的保留率为71.94%,并分离出柠檬桉树脂多糖1(ERP1)、柠檬桉树脂多糖2(ERP2)。以抗坏血酸(Vc)作参照物,通过对DPPH·自由基和·OH自由基清除实验,考察了ERP的抗氧化活性。结果表明,ERPC、ERP1、ERP2对DPPH·自由基和·OH自由基都有较明显的清除能力,且在一定的浓度范围内呈现出一定的剂量依赖性,三者对DPPH·自由基清除的IC_(50)值分别为9.82,13.6,11.3 mg/L,对·OH自由基清除的IC_(50)值分别为23.6,26.9,53.7 mg/mL,但均相比于同浓度下的Vc抗氧化活性较弱。Eucalyptus citriodora resin polysaccharide(ERP)was extracted by the water extraction and alcohol precipitation method,and the phenol-sulfuric acid method was used to determine the polysaccharide content,and Eucalyptus citriodora resin crude polysaccharide(ERPC)content was 7.79%.The Sevage method and the trichloroacetic acid method(TCA)were used to deproteinize ERP.The results show that the trichloroacetic acid method has a better effect on ERP deproteinization,and when the TCA concentration reaches 6%,the protein clearance rate reaches 94.79%,and polysaccharides are retained that rate is 82.99%.The polyamide chromatography column was used to depigment ERP,the decolorization rate was 77.95%,the polysaccharide retention rate was 71.94%,and ERP1 and ERP2 were separated.With ascorbic acid(Vc)as the reference material,the DPPH free radical and OH free radical scavenging experiment and the reduction ability determination experiment were passed.The antioxidant activity of ERP was investigated.The results showed that:ERPC,ERP1,and ERP2 have obvious scavenging ability on DPPH·radicals and·OH free radicals,and show a certain dose-dependent within a certain concentration range,and the three are effective for scavenging DPPH·radicals.The IC_(50) values were 9.82,13.6,11.3 mg/L,respectively,and the IC_(50) values for·OH radical scavenging were 23.6,26.9,53.7 mg/mL,respectively,but they were all weaker than the Vc antioxidant activity at the same concentration.
