摘 要:建立超高效液相色谱法(UPLC)测定黑木耳中麦角甾醇含量的方法,并将黑木耳麦角甾醇含量与性状特征和膨胀度进行相关性研究。测定条件:色谱柱如下,Waters ACQUITY UPLC BEH C色谱柱(100 mm 2.1 mm,1.7μm),流动相为乙腈:水(98:2)等度洗脱,流速:0.5 mL/min,柱温:35℃,进样量:2μL,检测波长:282 nm。结果显示麦角甾醇在0.0016~0.08 mg/mL范围内呈良好的线性关系,y=5514333.6315x+820.384(R~2=0.9999),平均回收率为100.89%,RSD值为0.55%。46份黑木耳样品中麦角甾醇含量在0.1359~0.8936 mg/g之间,平均含量为0.6075 mg/g,不同黑木耳的麦角甾醇含量差异显著(P<0.01),麦角甾醇含量与子实体厚度和膨胀度分别存在极显著正相关关系(P<0.01)。本研究对黑木耳进行科学的质量评价,为进一步研究、生产和应用黑木耳作为功能性食品和治疗剂提供研究基础。A method for the determination of ergosterol content in Auricularia heimuer by ultra performance liquid chromatography(UPLC)was established,and the correlation between the content of ergosterol in Auricularia heimuer and its characteristics and turgidity was studied.Test conditions as follows,chromatographic column:Waters ACQUITY UPLC BEH Ccolumn(100 mm 2.1 mm,1.7μm),mobile phase was acetonitrile:water(98:2)isocratic elution,flow rate was 0.5 mL/min,column temperature was 35℃,injection volume was 2μL,detection wavelength was 282 nm.The results show ergosterol in the 0.0016~0.08 mg/mL range showed a good linear relationship,y=5514333.6315x+820.384(R~2=0.9999)the average recovery was 100.89%,the RSD value was 0.55%.The ergosterol content of 46 Auricularia heimuer was between 0.1359~0.8936 mg/g,and the average content was 0.6075 mg/g.The ergosterol content of different Auricularia heimuer was significantly different(P<0.01)and there were highly significant positive correlations between ergosterol content and sporocarp thickness and turgidity(P<0.01)This study provides a scientific quality evaluation of Auricularia heimuer to provide a research basis for further research,production and application of Auricularia heimuer as a functional food and therapeutic agent.
