摘 要:淀粉的合成受一系列酶协同作用调控,明确块茎发育过程中淀粉合成相关基因的表达模式和淀粉颗粒粒径的变化规律,可以为马铃薯高淀粉育种提供分子基础.以块茎发育7个时期的样品为研究对象,用实时荧光定量PCR方法检测SuSy4等8个淀粉合成相关基因的表达量,用显微镜法测定淀粉颗粒粒径.结果表明,淀粉合成相关基因在块茎中高表达,在块茎发育时期,后期表达要高于前期表达.在发育过程中,小颗粒淀粉占比最大,大颗粒淀粉占比总体呈现出逐渐增加的趋势.Starch synthesis is coordinated by multiple enzymes.Analysis of starch synthesis related gene expression and starch granule size during developmental stages of potato tubers will provide a molecular basis for potato breeding aiming of high starch content.With the tuber samples of seven tuber developmental stages, we investigated the expression of eight starch synthesis related genes using quantitative reversetranscription PCR(RT-qPCR) method, and examined the granule size using microscopic method.The results showed that eight genes were preferentially expressed in tubers, and their transcript levels in the late stages were higher than that in the early stages of tuber development.The small starch granule with size < 20 μm made up of the majority starch granule, and the proportion of large granules with size ≥ 20 μm generally increased with the tuber development.
