摘  要:BMI-1(B-cell specific moloney leukemia virus insertion site 1,BMI-1)于1991年在荷兰癌症中心首次发现,是哺乳动物中发现的第一个多梳基因家族成员。其作为一种原癌基因,参与小鼠T、B细胞淋巴瘤的发生发展过程。多梳基因(ploycomb group genes,PCG)家族是以转录调控因子为主并且与发育相关的一组重要基因族,在生物生长发育过程中有着重要的作用。BMI-1基因与肿瘤关系之间的研究是近年来的研究热点,早期研究发现,BMI-1可参与调节肿瘤干细胞增殖、分化、衰老的过程,BMI-1在多种恶性肿瘤中表达上调,并且与肿瘤病理类型、临床期别、侵袭、转移、预后相关。最新研究表明,可通过多种途径抑制或沉默BMI-1基因的表达,进而抑制肿瘤细胞的增殖、分化过程,增强恶性肿瘤细胞对化疗及放疗的敏感性。这为今后寻找新的生物治疗靶点,提高患者生存率,改善患者预后提供了新的证据。本文就BMI-1基因的表达与各类型肿瘤发生发展、治疗、预后的关系及可能参与的信号途径作一综述。BMI-1 is a member of the polyphobic gene family.The ploycomb group genes family is an important group of genes that are mainly transcriptional regulators and related to development.The relationship between BMI-1 gene and tumor has been a hot topic in recent years.BMI-1 is involved in regulating the proliferation,differentiation and aging of tumor stem cells in early studies.BMI-1 is up-regulated in a variety of malignancies and is related to the pathological type,clinical stage,invasion,metastasis and prognosis of tumors.It is closely related to the occurrence and development of malignant tumors.Recent studies have shown that many approaches can inhibit the proliferation and differentiation of tumor cells to enhance the sensitivity of malignant tumor cells to chemotherapy and radiotherapy by inhibiting the expression of BMI-1 gene.This provides new evidence for the combination of biological targeted therapy and traditional therapy to treat malignant tumors,improve the survival rate of patients in the future.This paper reviews the relationship between the expression of BMI-1 gene and the development,treatment and prognosis of various types of tumors,as well as the targeted inhibitors of BMI-1 and the possible signaling pathways.
