摘 要:目的 探讨宫颈癌组织Irisin含量变化及其对肿瘤细胞增殖和侵袭的影响。方法 选择宫颈癌组织78例份、正常宫颈组织68例份,采用ELISA法检测Irisin含量,并分析宫颈癌组织Irisin含量与临床病理特征的关系。体外传代培养人宫颈癌细胞株HeLa(以下称HeLa细胞),随机分为Irisin组和对照组。Irisin组加入适量Irisin,使其终浓度为5 nmol/L,对照组不予干预,继续培养12 h,收集细胞,采用CCK-8法检测细胞增殖活性,采用Transwell侵袭实验检测细胞侵袭能力,采用Western blotting法检测MMP-2、MMP-9蛋白表达。结果 宫颈癌组织Irisin含量显著高于正常宫颈组织(P<0.05)。宫颈癌组织Irisin含量与FIGO分期、组织分化程度、淋巴结转移有关(P均<0.05),而与年龄、病理类型、浸润深度无关(P均>0.05)。Irisin组细胞增殖活性、侵袭能力以及MMP-2、MMP-9蛋白相对表达量均显著高于对照组(P均<0.05)。结论 宫颈癌组织Irisin含量显著升高,且其含量与FIGO分期、组织分化程度、淋巴结转移有关;Irisin能够增强宫颈癌细胞增殖活性和侵袭能力,其机制可能与促进MMP-2、MMP-9蛋白表达有关。Objective To investigate the expression changes of Irisin in the cervical cancer tissues and to explore its effects on the proliferation and invasion of tumor cells.Methods We selected 78 cervical cancer tissues and 68 normal cervical tissues.The content of Irisin was detected by ELISA,and we analyzed the relationship between the content of Irisin in cervical cancer tissues and the clinicopathological characteristics of the cervical cancer.The human cervical cancer cell line HeLa cells(hereinafter referred to as HeLa cells) were cultured in vitro and randomly divided into the Irisin group and control group,with 6 replicate wells in each group.In the Irisin group,we added Irisin to make the final concentration be 5 nmol/L.The control group was not intervened.Cells of these two groups were continued to be cultured for 12 h and then were collected.The cell proliferation activity was detected by CCK-8 method.Transwell invasion assay was used to measure the invasion of HeLa cells.The protein expression of MMP-2 and MMP-9 were detected by Western blotting.Results The expression of Irisin in the cervical cancer tissues were obviously higher than that in the normal cervical tissues(P<0.05),and the expression of Irisin in the cervical cancer tissues was related to FIGO staging,pathological grade,and lymph node metastasis(all P<0.05),but not related to age,pathological type,or depth of invasion(all P>0.05).The cell proliferation and invasion ability of the Irisin group were significantly higher than those of the control group(both P<0.05).The relative expression levels of MMP-2 and MMP-9 proteins in the Irisin group were significantly higher than those in the control group(both P<0.05).Conclusion The expression of Irisin in the cervical cancer tissues increases,its content may be related to the FIGO staging,pathological grade,and lymph node metastasis.Irisin could increase the proliferation and invasion abilities of HeLa cells perhaps by increasing the protein expression of MMP-2 and MMP-9.
