摘  要:目的探讨布托啡诺通过调控巨噬细胞极化对乳腺癌细胞侵袭和迁移的影响及其机制。方法MTT法检测RAW细胞和MCF-7细胞存活情况。Transwell检测MCF-7细胞侵袭和迁移数量;蛋白质免疫印迹(Western blot)检测RAW细胞M2型相关细胞Arg-1蛋白和MCF-7细胞MMP-2、MMP-9蛋白表达。结果与RAW组比较,M2RAW组细胞存活率和Arg-1蛋白表达水平显著升高(P<0.05);与M2RAW组比较,布托啡诺+M2RAW组细胞存活率和Arg-1蛋白表达水平显著降低(P<0.05)。与MCF-7组比较,布托啡诺+MCF-7组细胞存活率、侵袭、迁移细胞数和MMP-2、MMP-9蛋白表达显著降低(P<0.05)。与RAW+MCF-7组比较,M2RAW+MCF-7组细胞存活率、侵袭、迁移细胞数和MMP-2、MMP-9蛋白表达显著升高(P<0.05);与M2RAW+MCF-7组比较,布托啡诺+M2RAW+MCF-7组细胞存活率、侵袭、迁移细胞数和MMP-2、MMP-9蛋白表达显著降低(P<0.05)。结论布托啡诺通过抑制巨噬细胞M2型极化从而抑制乳腺癌细胞侵袭和迁移。Objective To investigate the effect of butorphanol on breast cancer cell invasion and migration and its mechanism by regulating macrophage polarization.Methods The MTT method detects the survival of RAW cells and MCF-7cells.Transwell detects the number of invasion and migration of MCF-7 cells.Western blot detects Arg-1 protein in M2-type related cells of RAW cells and the protein expression of MMP-2 and MMP-9 in MCF-7 cells.Results Compared with the RAW group,the cell survival rate and Arg-1 protein expression level in the M2RAW group were significantly increased(P<0.05).Compared with the M2RAW group,the cell survival rate and Arg-1 protein expression level in the butorphanol+M2RAW group were significantly higher decrease(P<0.05).Compared with the MCF-7 group,the butorphanol+MCF-7group had significantly lower cell viability,invasion and migration cell numbers,and MMP-2 and MMP-9 protein expressions(P<0.05).Compared with the RAW+MCF-7 group,the cell survival rate,invasion and migration cell numbers,and MMP-2and MMP-9 protein expressions in the M2RAW+MCF-7 group were significantly increased(P<0.05).Compared with the butorphanol+M2RAW+MCF-7 group,the cell survival rate,invasion,migration cell number and MMP-2,MMP-9 protein expressions were significantly decreased(P<0.05).Conclusion Butorphanol inhibits the invasion and migration of breast cancer cells by inhibiting the M2 polarization of macrophages.
