摘  要:目的探讨使用多聚赖氨酸-超微、超顺磁氧化纳米铁颗粒蛋白复合物-人胎盘间充质干细胞(PLL-USPION-PMSCs)分子探针靶向结直肠癌小鼠模型,观察活体行分子成像的可行性及PMSCs对结直肠癌的影响。材料与方法培养PMSCs,以PLL进行转染,用USPION对PMSCs-PLL进行磁性标记。将PLL-USPION-PMSCs分子探针通过鼠尾静脉移植入实验组(10只)结直肠癌动物模型内,以同样方法在荷瘤小鼠瘤内注入不含细胞的PBS作为空白对照(对照组,10只)。采用T2WI、T2mapping、T2*mapping序列采集注射后1、3、7、10和14 d两组荷瘤小鼠的MRI图像,观察两组移植瘤内信号分布、变化特征,测量肿瘤的大小、T2值、T2*值,绘制生长曲线。肿瘤标本称瘤重,计算抑瘤率,行普鲁士蓝染色、HE染色及免疫组化病理检测,进行统计学分析。结果实验组移植瘤内可检测到PMSCs低信号,且PMSCs信号和肿瘤信号及T2、T2*值随时间推移发生动态变化。两组移植瘤T2、T2*值差异有统计学意义(F=24.472、8.963,P=0.00、0.01)。两组移植瘤生长曲线相似且体积变化差异有统计学意义(F=4.32,P<0.05),抑瘤率为正值。实验组普鲁士蓝染色后红色肿瘤组织边缘区域可见PMSCs蓝染颗粒,HE染色示两组移植瘤新生血管丰富,免疫组化示两组移植瘤血管内皮标志物CD31、CD34均(+),按照Weidner的判断标准计数,血管数量差异均无统计学意义(t=0.245、0.270,P>0.05)。结论3.0T MRI可安全、有效地实现对PMSCs活体示踪,PMSCs对结直肠癌有定向趋化作用,可以聚集至肿瘤内,通过不同时期的促进和抑制肿瘤生长相互交叉作用最终抑制肿瘤生长。Purpose To explore the feasibility of molecular imaging of polylysine-ultramicro and superparamagnetic oxide nano-iron particle protein complexes and human placental mesenchymal stem cells(PLL-USPION-PMSCs)targeting colorectal cancer(CRC)tumorbearing mice and the effect of PMSCs on CRC.Materials and Methods The PMSCs were cultured,transfected with PLL,and PMSCs-PLL were magnetically labeled with USPION,the PLL-USPION-PMSCs molecular probe was transplanted into the experimental group(n=10)of the CRC animal model through the tail vein of the rat,in the same way,PBS without cells was injected into the tumor of the tumor-bearing mice as a blank control(n=10).The T2WI,T2mapping,T2*mapping sequences were used to observe the signal distribution and change characteristics of transplanted tumor and PMSCs at 1,3,7,10 and 14 days after injection.The size,T2,T2*value of tumor,calculated inhibition rate of all transplanted tumors were measured,and prussian blue,HE staining and immunohistochemical pathological detection,and statistical analysis was further performed.Results In the experimental group,the low signal of PMSCs could be detected in the transplanted tumor,and the signal of PMSCs and tumor as well as T2 and T2*values changed dynamically over time,and there were statistical differences in T2 and T2*values between the two groups(F=24.472,8.963,P<0.05).The growth curves of transplanted tumors in the two groups were similar and there were statistical differences in volume change between the two groups(F=4.32,P<0.05),and the inhibitory rate was positive.PMSCs blue-stained particles were found in the marginal area of the red tumor tissue after prussian blue staining in the experimental group,HE staining showed that the transplanted tumors of two groups were rich in new blood vessels.The vascular endothelial markers including CD31 and CD34 were positive in the transplanted tumors of two groups.According to Weidner's judgment standard,there was no statistical difference in vascular endothelial markers between the two groups(
