摘 要:目的:用基于L(3^(4))正交试验设计的指标成分含量关联生物活性的综合评分法确定脑脉通的最佳提取工艺。方法:利用高效液相色谱(HPLC)建立脑脉通有效成分测定方法,采用DPPH法和ABTS法对其进行体外抗氧化研究,采用生色底物法进行体外抗凝活性研究;以有效成分转移率、DPPH和ABTS自由基清除率和凝血酶抑制率作为指标,采用L(3^(4))正交试验设计优化脑脉通提取工艺,运用综合评分法确定最优工艺。结果:脑脉通的最佳提取工艺为:加入10倍量的60%乙醇进行加热回流提取2次,每次60 min。结论:优选的提取工艺合理可靠;多指标成分含量测定关联生物活性分析为中药产品制备工艺优化提供了方法借鉴。Objective: To determine the optimal extraction technology of Naomaitong through the comprehensive scoring method of “multiple index component-biological activity” based on L(3^(4)) orthogonal design. Method: The determination method of the active components in Naomaitong was established by HPLC. In vitro antioxidant study was conducted by DPPH and ABTS methods, and chromogenic substrate assay was used for in vitro anticoagulant activity study. Using the transfer rate of active constituents, DPPH and ABTS radical scavenging rate and thrombin inhibition rate as the indicators, the extraction technology of Naomaitong was optimized by the L(3^(4)) orthogonal design, and the comprehensive scoring method was used to determine the optimal technology. Result: The optimal extraction technology of Naomaitong was as follows: adding 10 times the amount of 60%ethanol for heating reflux extraction 2 times, each time for 60 min. Conclusion: The optimized extraction technology is reasonable and reliable. “Multiple index component-biological activity” provides the reference for the preparation process optimization of traditional Chinese medicine products.
