摘 要:花序大小是评价绣球花观赏性的重要指标,通常用花序直径来衡量植物花序的大小。全基因组关联研究(genome-wide association study,GWAS)能够识别与特定性状相关联的分子标记;利用这些分子标记可以进行分子标记辅助育种(molecular marker-assisted breeding,MAB),从而加快新品种的选育。本研究以125份绣球植株为材料,基于基因组重测序获得的SNP分子标记,利用混合线性模型(mixed linear model,MLM)进行全基因组关联分析,定位控制花序大小的关键基因。结果表明,通过GWAS分析检测到24个与花序大小相关的SNP位点[-log_(10)P≥5],表型贡献率在5.01%~23.48%之间;对定位的24个SNP位点进行基因注释,获得了74个候选基因,其中68个基因位于基因间隔区,6个位于基因编码区。在74个候选基因中57个具有基因功能注释,另外17个未能注释。根据注释结果,获得了6个与花序大小发育相关的基因,包括3个植物生长素响应基因(SAUR)、1个扩张蛋白基因、1个吲哚-3-乙酸诱导蛋白基因(ARG7)以及1个ABC转运蛋白基因。Inflorescence size,usually described as inflorescence diameter,is an important index to evaluate the ornamental value of Hydrangea macrophylla.Molecular markers associated with specific traits can be identified by genome-wide association study(GWAS).Using the molecular markers,molecular marker-assisted breeding(MAB) can be performed and greatly shorten the breeding period of new cultivars.In this article,utilizing 125 H.macrophylla plants as materials,the genome-wide association analysis,based on the SNP markers detected by genome resequencing,was carried out by mixed linear model(MLM) to locate the key genes controlling inflorescence size.The results showed that 24 SNP loci related to inflorescence diameter were detected by GWAS analysis [-log_(10)P≥5],and the phenotypic contribution rate ranged from 5.01% to 23.48%.Among the 24 SNPs loci,74 potential genes were obtained,of which 68 genes were located in the gene spacer regions,and the other 6 in the coding regions.Among these genes,57 were annotated,and the other 17 genes without any annotation.According to the annotation results,6 genes associated with inflorescence diameter were screened,including 3 auxin response genes SAUR,one expansin gene,one indole-3-acetic acid-induced protein ARG7 and one ABC transporter gene.
