摘  要:目的研究给予经典方剂当归补血汤或当归后兔血浆中阿魏酸药动学参数的变化规律。方法采用RP-HPLC测定血药浓度,以乙腈-水(含0.6 mol/L醋酸)[25:75(v/v)]为流动相,DIRMA Diamonsil C_(18)柱(200 mm×4.6 mm,5μm),紫外检测波长316 nm,流速1.0 mL/min。结果给予当归补血汤和当归后兔浆中阿魏酸的T_(max)、分别为(6.667±0.887)min和(8±0.992)min,AUC_(0-t)分别为(95768.726±14361.123)μg/L·min和(89161.107±12042.231)μg/L·min,MRT_(0-t)分别为(69.952±10.272)min和(56.106±7.399)min,C_(max)分别为(1485.929±202.052)μg/L和(1723.187±255.607)μg/L。结论给予当归补血汤后即黄芪、当归按5∶1比例配伍时阿魏酸的达峰时间提前,生物利用度增加,提示在当归补血汤中当归、黄芪按1∶5比例配伍时可保证药效充分发挥,为探讨经典方剂的组方规律提供了依据。Objective To study the pharmacokinetics of ferulic acid in rabbits plasma which were administrated with Danggui Buxue Decoction(当归补血汤)or Danggui(Angelicae Sinensis Radix).Methods The concentration of ferulic acid in plasma was analyzed by RP-HPLC with the mobile phase of acetonitrile-water(0.6 mol/L acetic acid)=25:75(v/v)and with the column of DIKMA Diamonsil C18(200 mm×4.6 mm,5μm),λ=316 nm.The flow rate was 1.0 mL/min.Result Administrated with Danggui Buxue Decoction or Danggui(Angelicae Sinensis Radix),the ferulic acid in rabbits plasma showed T_(max) was(6.667±0.887)min and(8±0.992)min,AUC_(0-t) was(95768.726±14361.123)μg/(L·min)and(89161.107±12042.231)μg/(L·min),MRT_(0-t) was(69.952±10.272)min and(56.106±7.399)min,C_(max) was(1485.929±202.052)μg/L and(1723.187±255.607)μg/L,respectively.Conclusion The peak time of ferulic acid was advanced and the bioavailability was increased after Danggui Buxue Decoction was given[Huangqi(Astragali Radix)and Danggui(Angelicae Sinensis Radix)were mixed at the ratio of 5∶1],suggesting that the compatibility of Huangqi(Astragali Radix)and Danggui(Angelicae Sinensis Radix)in Danggui Buxue Decoction at the ratio of 1∶5 can ensure the full play of the drug effect,providing a basis for exploring the composition rules of classical prescriptions.
