摘 要:目的探讨扶阳活血解毒方对溃疡性结肠炎大鼠的治疗作用及肠道菌群的影响。方法采用三硝基苯磺酸(TNBS)/乙醇法复制UC大鼠模型,随机分为正常对照组、模型组、扶阳活血解毒方高剂量组、扶阳活血解毒方中剂量组、扶阳活血解毒方低剂量组,每组10只。治疗14天后,观察大鼠一般状况并进行疾病活动指数(DAI)评分,HE染色观察结肠病理学变化,检测血清IL-4、IFN-γ的表达。结果与模型组相比,各治疗组DAI评分显著降低,结肠HE染色充血、水肿及炎性细胞侵润较模型组明显减轻,IFN-γ的水平降低,IL-4的水平升高,差距具有统计学意义(P<0.05)。结论扶阳活血解毒方对UC模型的大鼠症状及病理组织学有明显的改善作用,可能通过上调血清IL-4表达,减少IFN-γ的表达来维持TH1/TH2细胞平衡进而发挥治疗溃疡性结肠炎的作用。Objective To explore the therapeutic effect of Fuyang Huoxue Jiedu Recipe(扶阳活血解毒方)on ulcerative colitis(UC)rats and its influence on intestinal flora.Methods UC rat model was established by trinitrobenzene sulfonic acid(TNBS)/ethanol method and the rats were randomly divided into normal control group,model group,Fuyang Huoxue Jiedu Recipehigh,mediumandlow dose groups,with 10 rats in each group.After 14 days of treatment,the general condition of rats was observed,the disease activity index(DAI)was scored,the pathological changes of colon were observed by HE staining,and the expressions of serum interleukin 4(IL-4)and γ interferon(IFN-γ)was detected.Results Compared with those of the model group,the DAI score of each treatment group was significantly reduced,the congestion,edema and inflammatory cell invasion of colon were significantly reduced,the level of IFN-γ decreased and the level of IL-4 increased,and the difference was statistically significant(P<0.05).Conclusion Fuyang Huoxue Jiedu Recipe can obviously improve the symptoms and histopathology of UC model rats,which may maintain the balance of Th1/Th2 cells by up-regulating the expression of serum IL-4 and reducing the expression of IFN-γ,and then plays a role in treating ulcerative colitis.
