摘  要:以极具观赏价值的山楂资源为试材,探究不同取材部位、灭菌时间、不同培养基及植物生长调节剂对其组培快繁的影响,以建立山楂快繁体系。结果表明:将山楂带芽茎段用70%酒精灭菌30 s,0.1%Hg Cl2消毒8 min后,在启动培养基MS+0.5 mg/L 6-BA中进行启动培养,启动率达94.44%;在MS+0.2 mg/L 6-BA+0.02 mg/L NAA培养基中进行继代增殖,增殖系数达4.10;在1/2MS+0.5 mg/L IBA+0.2 mg/L NAA培养基中培养4 d后,转入1/2MS(蔗糖15 g/L)中进行生根培养,25 d后生根率达85.18%;移栽至纯蛭石基质中,成活率达92%。Using hawthorn resources with great ornamental value as test materials,this system aims to explore the effects of explant-providing organs,sterilization time,media and plant growth regulators on rapid propagation of hawthorn.The results showed that the initiation rate reached 94.44%in the medium MS+0.5 mg/L 6-BA after the bud stem of hawthorn was sterilized with 70%alcohol for 30 s and 0.1%Hg Cl2for 8 min;the proliferation rate reached 4.10 in the medium of MS+0.2 mg/L 6-BA+0.02 mg/L NAA;the rooting percentage reached 85.18%after it was in the medium of 1/2MS(sucrose 15 g/L)for 25 days,which had been before cultured in the medium of1/2MS+0.5mg/L IBA+0.2 mg/L NAA for 4 day;and the survival rate was 92%after it was transplanted into vermiculite.
