摘 要:目的 探讨聚(腺苷二磷酸-核糖)聚合酶-1[poly(ADP-ribose)polymerase-1,PARP-1]缺陷(PARP-1^(-/-))对苯并[a]芘(B[a]P)致肺损伤过程中表皮生长因子受体(epidermal growth factor receptor, EGFR)的调控。方法 以PARP-1小鼠和野生型小鼠为研究对象,每组40只,雌雄各半。实验组采用动式吸入染毒途径暴露10.0μg/m^(3)B[a]P,6 h/d, 5 d/周,对照组给予溶剂二甲基亚砜相同处理,暴露180天后,借助RT-PCR、Western blot和免疫荧光检测技术分析各组动物肺组织中表皮生长因子受体的表达变化。结果 与对照组相比,野生型小鼠B[a]P染毒后肺组织中的EGFR表达明显升高(P<0.05),PARP-1^(-/-)小鼠B[a]P染毒后肺组织中的EGFR激活受抑制(P<0.05)。结论 PARP-1缺陷可对抗B[a]P诱导的EGFR异常激活。OBJECTIVE To investigate the regulation of PARP-1 deficiency on epidermal growth factor receptor(EGFR) during lung injury of mice induced by benzo[a]pyrene(B[a]P) inhalation exposure.METHODS PARP-1 knockout mice(PARP-1^(-/-)) and WT mice were selected as the object, and which were randomly assigned into either an intervention or a control group(n=40, half male and half female). The intervention group were individually treated with 10.0 μg/m^(3)B[a]P for 180 days by dynamic inhalation exposure(6 h per day and 5 days per week), and the control group was given the solvent dimethyl sulfoxide(DMSO) during the same period. The expression of EGFR in lung tissues of animals were examined by RT-PCR, Western blot and immunofluorescence.RESULTS In WT mice, the intervention manifested significant increase expression of EGFR in lung tissue, but no changes were found in the control. In PARP-1mice, the intervention manifested significant inhibition expression of EGFR, but the control group exhibited no changes.CONCLUSION PARP-1^(-/-) deficiency suppresses the abnormal activation of EGFR during lung injury of mice induced by B[a]P inhalation exposure.
