摘  要:目的探讨“原络通经”针法调节痴呆小鼠肠道菌群多样性及肠道SCFA代谢的机制研究。方法SAMP8小鼠随机分成3组:模型对照组、原络通经组、百会针刺组,每组6只;以SAMR1同品系正常小鼠6只作为空白对照组,通过Morris水迷宫检测治疗后小鼠空间学习记忆能力的变化,免疫组织化学分析β淀粉样蛋白1-42(Aβ_(1-42))表达水平,qRT-PCR和Western blot检测Aβ_(1-42)和淀粉样蛋白前体蛋白(APP)表达情况,16S rRAN测序检测肠道菌群多样性,采用气相色谱质谱联用方法分析肠道短链脂肪酸(SCFA)。结果与空白对照组相比,模型对照组小鼠逃避潜伏时间显著增加,原络通经组和百会针刺组逃避潜伏时间显著减少,且原络通经组改善更显著(P<0.05);与空白对照组相比,模型对照组脑组织Aβ_(1-42)和APP基因表达显著增加,原络通经组和百会针刺组基因表达显著减少,且原络通经组改善更显著(P<0.05);与空白对照组相比,模型对照组肠道菌群多样性和肠道短链脂肪酸含量显著降低,原络通经组和百会针刺组肠道菌群多样性和肠道短链脂肪酸含量显著增高,且原络通经组改善更显著(P<0.05)。结论原络通经疗法与百会针刺均可改善痴呆小鼠学习记忆功能障碍,降低脑组织Aβ_(1-42)和APP基因表达,改善肠道菌群多样性及增加肠道短链脂肪酸含量,而原络通经疗法疗效更优。Objective To investigate the mechanism of“Yuanluo Tongjing”acupuncture method in regulating intestinal flora diversity and intestinal SCFA metabolism in demented mice.Methods SAMP8 mice were randomly divided into three groups:model control group,Yuanluo Tongjing group and Baihui acupuncture group,6 mice in each group;6 normal mice of the same strain of SAMR1 were used as blank control group.Aβ_(1-42) and APP protein expression,16S rRAN sequencing to detect intestinal flora diversity,and gas chromatography-mass spectrometry to analyze intestinal short-chain fatty acids(SCFA).Results Compared with the blank control group,the escape latency time was significantly increased in the model control group,and decreased in the Yuanluo Tongjing and Baihui acupuncture groups,and the improvement was more significant in the Yuanluo Tongjing group(P<0.05).Compared with the blank control group,the intestinal flora diversity and intestinal short-chain fatty acid content were significantly decreased in the model control group,while the intestinal flora diversity and intestinal short-chain fatty acid content were significantly increased in the Yuanluo Tongjing and Baihui acupuncture groups,and the improvement was more significant in the Yuanluo Tongjing group(P<0.05).Conclusion Both the Yuanluo Tongjing therapy and Baihui acupuncture can improve the learning and memory dysfunction,reduce the expression of Aβ_(1-42) and APP genes in brain tissue,improve the intestinal flora diversity and increase the intestinal short-chain fatty acid content in demented mice,and the Yuanluo Tongjing therapy is more effective.
