摘  要:旨在探究尿酸对液态保存猪精子质量的影响。通过精子分析仪检测精子活力,低渗肿胀法检测精子的质膜完整性、比色法和硫代巴比妥酸法分别检测总抗氧化能力和丙二醛含量,荧光探针染色检测顶体完整性、活性氧含量和线粒体膜电位,体外受精检测精子受精能力和胚胎发育能力。结果显示,在保存液中添加一定质量浓度的尿酸有助.于提高猪精子质量,同时发现尿酸主要是通过提高精子的抗氧化能力和线粒体膜电位来维持精子质量。此外,本研究发现尿酸还能够提高精子受精能力和胚胎发育能力。结果表明,添加5.0 mg/L尿酸对17℃液态保存的猪精液有利,可为新型猪精液保存液的研发提供依据。The purpose of this study is to explore the effect of uric acid on the quality of liquid-preserved pig sperm.Sperm motility was assessed by sperm analyzer, plasma membrane integrity was detected by hypotonic swelling method, total antioxidant capacity and malondialdehyde content were analysis by colorimetry and thiobarbituric acid method.Acrosome integrity, reactive oxygen species content and mitochondrial membrane potential were measured by fluorescent probe staining.The sperm fertilization ability and embryonic development ability were evaluated by in vitro fertilization.The results showed that adding a certain concentration of uric acid to the Modena extender could improve the quality of boar sperm.At the same time, it was found that uric acid mainly maintained the quality of sperm by improving the anti-oxidation ability of sperm and maintaining the mitochondrial membrane potential.In addition, this study found that uric acid could improve sperm fertilization and embryonic development.In summary, the addition of 5.0 mg/L uric acid is beneficial to boar semen stored in a liquid state at 17℃,and can provide experimental evidence for exploring a new type of semen preservation extender.
